Orphan nuclear receptor that can act as a repressor or activator of transcription. An important repressor of nuclear receptor signaling pathways such as retinoic acid receptor, retinoid X, vitamin D3 receptor, thyroid hormone receptor and estrogen receptor pathways. May regulate gene expression during the late phase of spermatogenesis. Together with NR2C1, forms the core of the DRED (direct repeat erythroid-definitive) complex that represses embryonic and fetal globin transcription including that of GATA1. Binds to hormone response elements (HREs) consisting of two 5'-AGGTCA-3' half site direct repeat consensus sequences. Plays a fundamental role in early embryonic development and embryonic stem cells. Required for normal spermatogenesis and cerebellum development. Appears to be important for neurodevelopmentally regulated behavior (By similarity). Activates transcriptional activity of LHCG. Antagonist of PPARA-mediated transactivation.

SEMA6A belongs to a subfamily characterized by an extracellular semaphorin domain, a transmembrane domain, and a long cytoplasmic tail. Members of this class can repel sympathetic and dorsal root ganglion axons in vitro, consistent with a traditional role as guidance signals. However, the length of the cytoplasmic tail, which includes an EVL-binding site in SEMA6A and an Src-binding site in SEMA6B, suggests that these semaphorins may also function as receptors. SEMA6A is expressed in developing neural tissue and is required for proper development of the thalamocortical projection. SEMA6A directly links the Ena/VASP and the semaphorin protein families since the SEMA6A protein is capable of selective binding to the protein EVL (Ena/VASP-like protein.

TCTN2 Antibody: TCTN2 is a member of the Tectonic protein family, a group of evolutionarily conserved secreted and transmembrane proteins that regulate the Hedgehog (Hh)-mediated patterning of the neural tube. While the expression pattern and function of TCTN2 is not known, it is 49% similar to TCTN1. TCTN1 is expressed during embryonic development in regions that participate in Hh signaling, beginning in the gastrulation stages in the ventral node. Mice expressing mutant TCTN1 die between E13.5 and E16.5 and display holoprosencephaly, a defect associated with reduced Hh signaling, indicating the role of TCTN1 as an Hh activator. At later stages in development, TCTN1 is thought to also act as a repressor on the Hh pathway in the anterior and posterior neural tube.

Ephrin B molecules expressed at the membrane surface bind to the Ephrin B family receptors on target cells during cell to cell contact. This interaction leads to cell signaling in the target cell but also generates a reverse signal in the cell expressing Ephrin B on its surface. This reverse signaling event is thought to be critical for vessel maturation and neuronal development. Importantly, tyrosine phosphorylation of Ephrin B is thought to be a critical component of this reverse signaling event. Recent work demonstrated that Tyr331 of Ephrin B was phosphorylated in HEK293 cells after stimulation by the soluble Ephrin B2 receptor tyrosine kinase.

T-cadherin Antibody: T-cadherin was initially identified as cadherin-type cell adhesion molecule expressed in various neuronal populations in a temporally and spatially restricted pattern during axon growth. T-cadherin is an atypical member of the cadherin family because it does not possess the typical transmembrane and cytoplasmic domains but is instead anchored to the plasma membrane by glycosylphosphatidylinositol (GPI) linkage. T-cadherin may play a role in malignant tumor development as loss of the chromosome locus containing the T-cadherin gene correlates with the development of a variety of cancers. Recently it has been shown that T-cadherin can act as a receptor for hexameric and high-molecular weight forms of adiponectin, suggesting that T-cadherin may also play a role in metabolic regulation.
