ICAM5 is a member of the intercellular adhesion molecule (ICAM) family. All ICAM proteins are type I transmembrane glycoproteins, contain 2-9 immunoglobulin-like C2-type domains, and bind to the leukocyte adhesion LFA-1 protein. This protein is expressed on the surface of telencephalic neurons and displays two types of adhesion activity, homophilic binding between neurons and heterophilic binding between neurons and leukocytes. It may be a critical component in neuron-microglial cell interactions in the course of normal development or as part of neurodegenerative diseases.

The MF23 monoclonal antibody specifically reacts with the mouse 50-55 kDa Foxp3 protein (JM2, IPEX), a member of the forkhead family of transcription factors. Foxp3 is expressed by the Treg lymphocytes, whose development and function are influenced by the forkhead protein. Ectopic expression of Foxp3 in T lymphocytes inhibits their activity and cytokine expression. Mutations of Foxp3 result in the “scurfy†mice phenotype. It is reported that the MF23 antibody recognizes an epitope between 1-87 amino acids in the N-terminal domain of mouse Foxp3.

It binds to FLT3 (CD135) which is found on multipotent progenitor (MPP) and common lymphoid progenitor (CLP) cells in mice. FLT3L induces the mobilization of the hematopoietic progenitors and stem cells in vivo which may help the system to kill cancer cells. FLT3L is crucial for steady-state pDC (plasmacytoid dendritic cells) and cDC (classical dendritic cells) development. Deficiency of FLT3L causes a dramatic decrease in DC numbers, whereas increasing its availability increase in DC numbers.

Girdin Antibody: Girdin is a Galpha-interacting protein that can enhance the activation of the protein kinase Akt, remodel the actin cytoskeleton, and is thought to be involved in the regulation of cell migration and cancer metastasis. It has recently been shown that Girdin interacts with Disrupted-in-Schizophrenia 1 (DISC1), a susceptibility gene for major psychiatric disorders. DISC1 is thought to be involved in the migration, positioning and differentiation of dentate granule cells (DGCs) during development; depletion of Girdin or blocking the Girdin-DISC1 interaction results in defects in axonal sprouting in the CA3 region of the hippocampus and overextended migration of mispositioning of DGCs, suggesting the Girdin plays a role in postnatal neurogenesis in the dentate gyrus.
