The 53-6.7 antibody reacts with the 32-34 kDa alpha subunit of mouse CD8, known as CD8a or CD8 alpha. CD8a can form a homodimer (CD8 alpha-alpha), but is more commonly expressed as a heterodimer with a second chain known as CD8b or CD8 beta. CD8 acts as a co-receptor in antigen recognition and subsequent T cell activation that is initiated upon binding of the T cell receptor (TCR) to antigen-bearing MHC Class I molecules. The cytoplasmic domains of CD8 provide binding sites for the tyrosine kinase lck, facilitating intracellular signaling events that lead to T cell activation, development, and cytotoxic effector functions. CD8+ cytotoxic T cells (CTLs) play an important role in inducing cell death of tumor cells, as well as cells infected by virus, bacteria or parasites.

Protein Kinase c alpha (PKC alpha) is an 77 kDa member of the conventional group (cPKCs: sensitive to calcium, diacylglycerol, phosphatidylserine and phorbol esters) of the PKC family of serine/ threonine kinases that are involved in a wide range of physiological processes including mitogenesis, cell survival and transcriptional regulation. PKC alpha is an ubiquitously expressed PKC isozyme that has been implicated in the regulation of a broad range of cellular functions including proliferation, differentiation, development, migration, cell cell adhesion, cell extracellular matrix adhesion, and solute transport. The activation loop threonine (threonine 497 in PKC alpha) of conventional PKCs is phosphorylated by phosphoinositide dependent kinase 1 (PDK1). This phosphorylation is necessary for the autophosphorylation of threonine 638 in the carboxy terminus of PKC alpha, a step that is critical for regulating the rate of PKC alpha dephosphorylation and inactivation.

SOCS1 Antibody: The Suppressor of cytokine signaling (SOCS) and cytokine-inducible SH2 proteins are a family of intracellular proteins which regulate the immune cell responses to cytokines. SOCS1 acts to suppress dendritic cell (DC) as well as T cell hyperactivation following cytokine signaling by inhibiting JAK tyrosine kinase, a kinase necessary for type I and II cytokine receptors to initiate signaling, by directly binding to the catalytic domain of the kinase. SOCS1 also possesses E3 ubiquitin protein ligase activity that results in the polyubiquitination of its target proteins and subsequent degradation by the proteosome. It is through this method that SOCS1 negatively regulates signaling by Toll-like receptors TLR2 and TLR4 by mediating the degradation of the TLR signaling adaptor protein TIRAP.
