The screening of cDNA expression libraries from human tumors with serum antibody (SEREX) has proven to be a powerful method for identifying the repertoire of tumor antigens recognized by the immune system of cancer patients, referred to as the cancer immunome. In this regard, cancer/testis (CT) antigens are of particular interest because of their immunogenicity and restricted expression patterns. Synoivial sarcomas are striking with regard to CT antigen expression, however, highly expressed in sarcoma, CT antigens do not induce frequent humoral immune responses in sarcoma patients. Sera from two patients were used to immunoscreen cDNA libraries from two synovial sarcoma cell lines and normal testis, resulting in the identification of 113 distinct antigens. Sarcoma antigen NY-SAR-91 is one of them.

Cluster of differentiation 10 (CD10) is also known as membrane metallo-endopeptidase, neutral endopeptidase (NEP), Neprilysin, and common acute lymphoblastic leukemia antigen (CALLA), is a 90-110-kDa type II transmembrane glycoprotein normally expressed by a variety of tissues, including epithelial cells of the prostate, kidney, intestine, endometrium, adrenal glands, and lung. This zinc-dependent metalloprotease enzyme cleaves peptide bonds on the amino side of hydrophobic residues and inactivates a variety of physiologically active secreted peptides. CD20 is thought to be the rate-limiting degrading enzyme of amyloid ? peptide (A?) whose abnormal misfolding and aggregation in neural tissue has been implicated in the development of Alzheimer's disease (AD). CD10 is also identified as the common acute lymphoblastic leukemia antigen (CALLA) present on leukemic cells of pre-B phenotype, and thus serves as the most important biomarker in the diagnosis of human acute lymphocytic leukemia (ALL).

Azurocidin is an Azurophil granule antibiotic protein, with monocyte chemotactic and antibacterial activity. The Azurophil granules, specialized lysosomes of the neutrophil, contain at least 10 proteins implicated in the killing of microorganisms. Azurocidin is a member of the serine protease family that includes Cathepsin G, Neutrophil Elastase (NE), and Proteinase 3 (PR3), however, Azurocidin is not a serine proteinase since the active site serine and histidine residues are replaced. Human Azurocidin together with NE and PR3 are expressed coordinately and are packaged together into azurophil granules during neutrophil differentiation. Azurocidin has been identified as a modulator of endothelial permeability and an important multifunctional inflammatory mediator. Neutrophils arriving first at sites of inflammation release Azurocidin which acts in a paracrine fashion on endothelial cells causing the development of intercellular gaps and allowing leukocyte extravasation. Azurocidin thus be regarded as a reasonable therapeutic target for a variety of inflammatory disease conditions.

Serpins are a group of proteins with similar structures that were first identified as a set of proteins able to inhibit proteases. They are the largest and most diverse family of serine protease inhibitors which are involved in a number of fundamental biological processes such as blood coagulation, complement activation, fibrinolysis, angiogenesis, inflammation and tumor suppression and are expressed in a cell-specific manner. Serpin E1 is a secreted protein which belongs to the Serpin family. Serpin E1 acts as 'bait' for tissue plasminogen activator, urokinase, and protein C. Its rapid interaction with TPA may function as a major control point in the regulation of fibrinolysis. Defects in SERPINE1 are characterized by abnormal bleeding due to Serpin E1 defect in the plasma. High concentrations of Serpin E1 have been associated with thrombophilia which is an autosomal dominant disorder in which affected individuals are prone to develop serious spontaneous thrombosis.

TSLPR is a type I membrane receptor that forms a functional heterodimeric complex with IL7R to bind TSLP. The TSLP R contains a WSXWS motif required for proper protein folding and a box1 motif important for association with the JAKs. TSLPR has a predicted molecular weight approximately 41 kD, and two further isoforms have been reported that are produced by alternative splicing. The TSLPR is expressed preferentially in myeloid cells including dendritic cells and activated monocytes, and is weakly expressed in T cells. Expression has also been reported in heart, skeletal muscle, and kidney tissues. TSLP binding to the heterodimeric functional receptor (TSLPR and IL7R) activates JAK2, STAT3 and STAT5 to stimulate cell proliferation. Ligand receptor interactions haves been implicated in the development of the hematopoietic system, dendritic cell maturation, and the maintenance and polarization of human Th2 memory T cells in allergic diseases.
