PLK1 (polo-like kinase 1) is a member of the serine/threonine protein kinase family, cdc5/polo subfamily. PLK1 contains two polo box domains with a predicted molecular weight of 68 kDa. PLK1 has been shown to regulate cdc2/cyclin B through phosphorylation and activation of cdc25c phosphatase. PLK1 is modified by phosphorylation at Threonine 210. PLK1 may also be required for cell division. Depletion of PLK1 results in apoptosis and deregulation of expression of PKL1 is correlated with development of many malignancies.

Limd1 (LIM domain-containing protein 1) is a ubiquitously expressed tumor suppressor containing 3 LIM zinc-binding domains. LIM domains consist of a cysteine-rich consensus sequence containing two distinct zinc-binding subdomains, which mediate protein-protein interactions. Limd1 interacts with the proteins SQSTM1, Rb, p62 and TRAF6. Limd1 was first identified when the deletion of its gene was noted in some cervical cancers. Limd1 blocks in vitro and in vivo tumor growth and is down-regulated in lung cancer. Limd1 may regulate osteoclast development under stressful conditions via its interactions with TRAF6 and p62.

DEAD-box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp, are putative RNA helicases implicated in several cellular processes involving modifications of RNA secondary structure and ribosome/spliceosome assembly. Based on their distribution patterns, some members of this family may be involved in embryogenesis, spermatogenesis, and cellular growth and division. DDX32 is a 743 amino acid nuclear protein that localizes to the mitochondria and is a member of the DEAD box helicase family. Expressed in various tissues, DDX32 is up-regulated by ionomycin in T lymphocytes and down-regulated in acute lymphoblastic leukemia. Considered a novel RNA helicase, DDX32 may play an important role in the development of colorectal cancer and may be involved in regulating T-cell response to certain apoptotic stimuli.

Beclin-1 Antibody: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. Beclin-1, a coiled-coil Bcl-2-interacting protein homologous to the yeast autophagy gene apg6, is a mammalian autophagy gene that can inhibit tumorigenesis and is expressed at reduced levels in human breast carcinoma, suggesting that defects in autophagy proteins may contribute to the development or progression of tumors. Bcl-2 can bind to Beclin-1 and inhibit Beclin-1-dependent autophagy in yeast and mammalian cells, suggesting that Bcl-2 functions as an anti-autophagy protein as well as an anti-apoptotic protein, which helps maintain autophagy at levels that are more compatible with cell survival rather than cell death.
