MAP kinases, also known as extracellular signal-regulated kinases (ERKs), act as an integration point for multiple biochemical signals. This protein kinase lies upstream of MAP kinases and stimulates the enzymatic activity of MAP kinases upon wide variety of extra- and intracellular signals. This gene is a member of the Src family of protein tyrosine kinases (PTKs). The encoded protein is a key signaling molecule in the selection and maturation of developing T-cells. It contains N-terminal sites for myristylation and palmitylation, a PTK domain, and SH2 and SH3 domains which are involved in mediating protein-protein interactions with phosphotyrosine-containing and proline-rich motifs, respectively. The protein localizes to the plasma membrane and pericentrosomal vesicles, and binds to cell surface receptors, including CD4 and CD8, and other signaling molecules. Multiple alternatively spliced variants, encoding the same protein, have been described.

The UCHT1 antibody is specific for human CD3e, also known as CD3 epsilon, a 20 kDa subunit of the T cell receptor complex, along with CD3 gamma and CD3 delta. These integral membrane protein chains assemble with additional chains of the T cell receptor (TCR), as well as CD3 zeta chain, to form the T cell receptor – CD3 complex. Together with co-receptors CD4 or CD8, the complex serves to recognize antigens bound to MHC molecules on antigen-presenting cells. These interactions promote T cell receptor signaling (T cell activation), inducing cell proliferation, differentiation, production of cytokines or activation-induced cell death. CD3 is differentially expressed during thymocyte-to-T cell development and on all mature T cells.

The RM4-5 monoclonal antibody specifically reacts with mouse CD4, also known as L3T4, a 55 kDa differentiation antigen expressed by the majority of thymocytes, subpopulations of mature T cells (like major histocompatibility complex class II-restricted T lymphocytes), a subset of natural killer T cells, and on pluripotent hematopoietic stem cells. CD4 binds to the major histocompatibility complex class II (MHC class II) and enhances T lymphocyte development and mature T cells functions. In T lymphocytes, CD4 binds to the cytoplasmic tail of enzyme tyrosine kinase (p56lck).Binding of RM4-5 is blocked by the anti-mouse CD4 clone GK1.5.
