Description:
The 1316H monoclonal antibody binds to mouse IL-13. IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and also by mast cells and NK cells. Targeted deletion of IL-13 in mice resulted in impaired Th2 cell development and indicated an important role for IL-13 in the expulsion of gastrointestinal parasites. IL-13 exerts anti-inflammatory effects on monocytes and macrophages and it inhibits the expression of inflammatory cytokines such as IL-1β, TNF-α, IL-6 and IL-8. IL-13 has also been shown to enhance B cell proliferation and to induce isotype switching, resulting in increased production of IgE. Blocking of IL-13 activity inhibits the pathophysiology of asthma. The 1316H antibody is reported to be a neutralizing antibody.

Translocation of portions of chromosome 8 with amplifications of the c-Myc gene are found in some leukemias and lymphomas, and typically associated with a poor prognosis. Portions of chromosome 8 have been linked to schizophrenia and bipolar disorder. Trisomy 8, also known as Warkany syndrome 2, most often results in early miscarriage but is occasionally seen in a mosaic form in surviving patients who suffer to a varying degree from a number of symptoms including retarded mental and motor development, and certain facial and developmental defects. WRN is a DNA helicase encoded by chromosome 8 and shown defective in those with the early aging disorder Werner syndrome. Chromosome 8 is also associated with Pfeiffer syndrome, congenital hypothyroidism and Waardenburg syndrome.

TEM1 Antibody: Tumor endothelial marker (TEM) 1 was originally identified as a human embryonic fibroblast-specific antigen and was later determined to be endosialin, a single-pass transmembrane glycoprotein that has multiple extracellular domains, including three EGF-like domains, a sushi-like domain, and a C lectin-like domain. TEM proteins are significantly up-regulated during angiogenesis and neoangiogenesis that are crucial for the growth of solid tumors. While TEM1 is not required for angiogenesis during fetal development, postnatal growth or wound healing, it plays a role in tumor growth, invasion, and metastasis. Fibronectin and collagen types I and IV act as specific ligands of TEM1, leading to suggestions that these molecules may cause changes in the extracellular matrix, cell adhesion and migration during tumor invasion.

CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 β chain (CD8 α/β) or as a homodimer (CD8 α/α). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed. CD8 is also found in littoral cell angioma of the spleen.

Epstein-Barr virus-induced gene 3 (Ebi3) is a widely expressed homolog to the interleukin IL-12 p40 subunit protein that forms a heterodimer with either IL-12 p35 or an IL-12 p35 homolog, p28, to create a new cytokine (IL-27). Ebi3 may function to antagonize IL-12 and to inhibit the development of a Th1 immune response. Ebi3 is strongly expressed in Hodgkin and Reed-Sternberg cells, independently of the EBV status of the tumor cells. Ebi2 is expressed in B-lymphocytes and lymphoid tissues and may function in the modulation of the immune system. Out of the nine genes that are induced by the Epstein-Barr virus, Ebi2 exhibits the highest levels of up-regulation. In addition, Ebi2 may play a role mediating normal lymphocyte functions.

CD55 is a member of the RCA (regulators of complement activation) family. RCA proteins is characterized by the presence of four to 30 SCRs (short consensus repeats also called CCPs for control protein modules) in their plasmaexposed regions. CD55 containing four SCR modules is involved in the regulation of the complement cascade. CD55 is known to bind CD97 via the first SCR. It also binds physiologically generated C3 convertases with its second and third SCRs. Binding results in an accelerated “decay”, or dissociation of active C3 convertases, thus blocking the development of C’ attack complexes on nonforeign cells. It is known that viruses and bacteria also utilize multiple SCR sites for infection.

CXCR4, a Chemokine Receptor involved in organ vascularization, neuronal cell migration, and patterning of the central nervous system during development. It binds stromal cell-derived factor 1 (SDF1, also called PBFS) and mediates migration of resting leukocytes and is unique in homing hematopoietic progenitors to bone marrow. These properties suggest that CXCR4 is involved in tumor cell migration and local tumor invasion. It is also an HIV-1 fusion co-factor that allows HIV-1 invasion in diverse human cell types. Although this receptor was initially called Neuropeptide Y3 Receptor, it does not respond to neuropeptide Y. CXCR4 has two isoforms that are produced by alternative splicing.
