Bone morphogenetic proteins (BMPs) constitute a subfamily within the TGF-beta superfamily of structurally related signaling proteins. Members of this superfamily are widely distributed throughout the body, and are involved in diverse physiological processes during both pre- and postnatal life. Like BMP-7, BMP-4 is involved in the development and maintenance of bone and cartilage. Description:
Members of the glial cell line-derived neurotrophic factor (GDNF) family, including GDNF and Neurturin, play key roles in the control of vertebrate neuronal survivial and differentiation. GDNF is a glycosylated, disulfide-bonded homodimer that is distantly related to the TGF superfamily of growth factors. Three receptors for these factors, GFR alpha-1, GFR alpha-2, and GFR alpha-3 have been identified. The receptors do not contain transmembrane domains and are attached to the cell membrane by glycosyl-phosphoinositol linkage. Both GFR alpha-1 and GFR alpha-2 have been shown to mediate the GDNF-dependent and Neurturin-dependent phosphorylation and activation of the tyrosine kinase Ret. GFR-3 is expressed only during development. GFR alpha-2 binds Neurturin and mediates activation of RET receptor tyrosine kinase by both Neurturin and GDNF.

ZNF536 Antibody: ZNF536 is a recently identified zinc-finger protein that is expressed primarily in the developing nervous system and the cerebral cortex, hippocampus, and hypothalamus. ZNF536 possess ten zinc fingers and interacts with CtBP1, a corepressor for gene transcription. It is most closely related to transcriptional repressor ZNF219. Overexpression of ZNF536 in embryonic stem cells dramatically reduced the mRNA levels of neuronal marker genes such as Pax6, MAP2, and beta-tubulin III following retinoic acid (RA)-induced differentiation, while depletion of ZNF536 via RNAi resulted in elevated mRNA levels of these genes, indicating its role in inhibiting neuronal cell differentiation. Overexpression of RA receptor a rescues the inhibitory role of ZNF536, suggesting that ZNF536 might inhibit RA response element-mediated transcriptional activity.

RAIDD Antibody: Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand of the TNF family through their death domain (DD)-containing receptors, TNFR1 and Fas. The death signals are transduced by a group of DD-containing adapter molecules. A novel cell death adapter was recently identified by two independent groups and designated RAIDD (RIP-associated ICH-1/CED-3-homologous protein with DD) and CRADD (caspase and RIP adapter with DD)1, RAIDD contains a DD and a CARD (for caspase recruitment domain) which interact with RIP and caspase, respectively, to transduce death signals. RAIDD is constitutively expressed in many tissues and mediates apoptosis caused by Fas and TNFR-1.

The GARP5 antibody reacts with human GARP, also known as LRRC32 or Garpin, an 80 kDa transmembrane protein which acts as a receptor for the latent form of TGF-beta 1 (pro-TGF-beta), preventing its secretion. Specifically, GARP is reported to associate with the pro-domain of TGF-beta 1, known as latency-associated peptide (LAP), which must be cleaved to release the biologically active cytokine. The putative role of GARP in sequestering the TGF-beta is important for regulating its activity, as TGF-beta signaling is involved in development and function of Th17, Treg and many other immune cell types. GARP expression has been shown on platelets and at high levels on Foxp3+ Treg cells, where it is proposed to be a phenotypic identifier for activated human Treg cells.

Existing as two alternatively spliced isoforms, C10orf27 is widely expressed in multiple tissues, including brain, thymus and testis. C10orf27 may be associated with multiple sclerosis (MS) susceptibility and pathogenesis. MS is an inflammatory disease that causes gradual destruction of myelin in the central nervous system. C10orf27 is encoded by a gene located on human chromosome 10, which contains over 800 genes and 135 million nucleotides, making up nearly 4.5% of the human genome. PTEN is an important tumor suppressor gene located on chromosome 10 and, when defective, causes a genetic predisposition to cancer development known as Cowden syndrome.

Chromosome 9 consists of about 145 million bases, represents 4% of the human genome and encodes nearly 900 genes. Thought to play a role in gender determination, deletion of the distal portion of 9p can lead to development of male to female sex reversal, the phenotype of a female with a male X,Y genotype. Hereditary hemorrhagic telangiectasia, which is characterized by harmful vascular defects, is associated with the chromosome 9 gene encoding endoglin protein, ENG. Familial dysautonomia is also associated with chromosome 9 though through the gene IKBKAP. Notably, chromosome 9 encompasses the largest interferon family gene cluster. CD19 is a transmembrane glycoprotein that contains two extracellular immunoglobulin-like domains. CD19 is present in both benign and malignant B-cells and is considered to be the most reliable surface marker of this lineage over a wide range of maturational stages. In normal lymphoid tissue, CD19 is observed in germinal centers, in mantle zone cells, and in scattered cells of the inter-follicular areas. Anti-CD19 exhibits an overall immunoreactivity pattern similar to those of the antibodies against CD20 and CD22. However, in contrast to CD20, expression of CD19 is continuous throughout B-cell development and through terminal differentiation of B-cells into plasma cells. Anti-CD19 positivity is seen in the vast majority of B-cell neoplasms commonly at a lower intensity than normal B-cell counterparts. Plasma cell neoplasms are nearly always negative, as are T-cell neoplasms.

Specificity of this MAb is validated by sequential immunoprecipitation with a PAb against Tenascin C. Tenascin C is a multifunctional, disulfide-linkedhexameric extracellular matrix glycoprotein expressed in association with mesenchymal epithelial interactions during development and in the neo-vasculature and stroma of undifferentiated tumors. In adults, it is restricted to certain epithelial-stromal interfaces and increases markedly in hyper-proliferative diseases and in stroma of many neoplasms, including gliomas, breast, squamous and lung carcinomas.
