Description:   SPG15 Antibody: Hereditary spastic paraplegias (HSPs) are genetically and phenotypically heterogeneous disorders.  Spastic paraplegia with thinning of the corpus callosum (ARHSP-TCC) is a relatively frequent form of complicated hereditary spastic paraplegia in which mental retardation and muscle stiffness at onset are followed by slowly progressive paraparesis and cognitive deterioration. SPG15 is the second gene known to be responsible for ARHSP-TCC in the Italian population. Mutations in this gene are associated with autosomal recessive spastic paraplegia-15. SPG15 encodes a protein containing a FYVE zinc finger binding domain which is thought to target these proteins to membrane lipids through interaction with phospholipids in the membrane. SPG15 mRNA is widely distributed in human tissues, as well as in rat embryos, suggesting a possible role for this protein during embryonic development. SPG15 co-localizes partially with endoplasmic reticulum and endosome markers, suggesting a role in intracellular trafficking.

Specificity of this MAb is validated by sequential immunoprecipitation with a PAb against Tenascin C. Tenascin C is a multifunctional, disulfide-linkedhexameric extracellular matrix glycoprotein expressed in association with mesenchymal epithelial interactions during development and in the neo-vasculature and stroma of undifferentiated tumors. In adults, it is restricted to certain epithelial-stromal interfaces and increases markedly in hyper-proliferative diseases and in stroma of many neoplasms, including gliomas, breast, squamous and lung carcinomas.

Description:   This gene encodes a 105 kD protein which can undergo cotranslational processing by the 26S proteasome to produce a 50 kD protein. The 105 kD protein is a Rel protein-specific transcription inhibitor and the 50 kD protein is a DNA binding subunit of the NF-kappa-B (NFKB) protein complex. NFKB is a transcription regulator that is activated by various intra- and extra-cellular stimuli such as cytokines, oxidant-free radicals, ultraviolet irradiation, and bacterial or viral products. Activated NFKB translocates into the nucleus and stimulates the expression of genes involved in a wide variety of biological functions. Inappropriate activation of NFKB has been associated with a number of inflammatory diseases while persistent inhibition of NFKB leads to inappropriate immune cell development or delayed cell growth. Description:   SnoN Antibody: TGF-beta is a ubiquitously expressed cytokine that signals through the Smad protein family to regulate numerous cellular processes such as embryonic development and tumorigenesis. This signaling is negatively regulated by Ski, the mammalian homolog of the transforming protein of an avian retrovirus that induces oncogenic transformation of chicken embryo cells, and the related protein SnoN. Like Ski, SnoN functions by binding to the Smad proteins and preventing their phosphorylation, thereby inhibiting their ability to bind DNA and activate the transcription of downstream genes. SnoN is located primarily in the nucleus in cancer tissues or cells, but in the cytoplasm in normal tissues or primary epithelial cells. There are at least four alternately spliced isoforms of SnoN; SnoN antibody will recognize all isoforms (SnoN, SnoN2, SnoI, and SnoA).

Description:   ARMER Antibody: Apoptosis is important for normal development and tissue homeostasis. It is mediated by various caspases and ultimately results in the activation of endogenous endonucleases that degrade cellular DNA. Although apoptosis induced by endoplasmic reticulum (ER) stress is thought to be mediated by caspase-12, other caspases such as caspase-9 are also thought to be activated following ER stress. Recently, ARMER, a novel integral ER-membrane protein was shown to protect cells from ER stress-induced apoptosis. Analysis of the caspase proteolytic cascade suggests that ARMER acts by inhibiting caspase-9 activity, although the mechanism for this remains unkown. It should be noted that ARMER is not related to the inhibitor of apoptosis proteins (IAP) family and does not contain any baculoviral IAP repeat (BIR) domains.

Description:   The PVM13-1 monoclonal antibody bings to human IL-13. IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and also by mast cells and NK cells. Targeted deletion of IL-13 in mice resulted in impaired Th2 cell development and indicated an important role for IL-13 in the expulsion of gastrointestinal parasites. IL-13 exerts anti-inflammatory effects on monocytes and macrophages and it inhibits the expression of inflammatory cytokines such as IL-1β, TNF-α, IL-6 and IL-8. IL-13 has also been shown to enhance B cell proliferation and to induce isotype switching, resulting in increased production of IgE. Blocking of IL-13 activity inhibits the pathophysiology of asthma. Cell death signals are transduced by death domain (DD), death effector domain (DED), and caspase recruitment domain (CARD) containing molecules. CARD containing proteins include some caspases, Apaf-1, CARD4, IAPs, RICK, ARC, RAIDD, BCL-10, and ASC. A novel CARD-containing protein was recently identified and designated CARD8.
