Programmed Death-1 (PD-1, CD279) is a type I transmembrane protein belonging to the CD28/CTLA-4 family of immunoreceptors that mediate signals for regulating immune responses. Members of the CD28/CTLA-4 family have been shown to either promote T cell activation (CD28 and ICOS) or downregulate T cell activation (CTLA-4 and PD-1). CD279 is expressed on activated T-cells, B-cells, myeloid cells and on a subset of thymocytes. In vitro, ligation of CD279 inhibits TCR-mediated T-cell proliferation and production of IL-1, IL-4, IL-10 and IFN-gamma. In addition, CD279 ligation also inhibits BCR mediated signaling. CD279 deficient mice have a defect in peripheral tolerance and spontaneously develop autoimmune diseases.

Mcl-1 Antibody: Myeloid cell leukimia-1 (Mcl-1) is a member of the Bcl-2 family of proteins that can act to promote cell survival. While the mechanism by which Mcl-1 inhibits apoptosis is not known, it is thought that it may heterodimerize and neutralize pro-apoptotic members of the Bcl-2 family such as Bim or Bak. Mcl-1 was originally identified in differentiating myeloid cells, but has since been shown to be expressed in multiple cell types. Mcl-1 is essential for embryogenesis and for the development and maintenance of B and T lymphocytes in animals. Mcl-1 exists as at least two distinct isoforms designated Mcl-1L and Mcl-1S. In marked contrast to the larger isoform of Mcl-1, overexpression of Mcl-1S promotes cell death.

The GK1.5 monoclonal antibody specifically binds with the mouse CD4 molecule, also known as L3T4, a 55 kDa differentiation antigen which binds to the MHC class II. CD4 is expressed on most thymocytes, a subpopulation of mature T lymphocytes, dendritic cells, pluripotent hematopoietic stem cells, B cell precursors, and lymphoid precursors inside the thymus. It is also expressed on the mouse egg cell membrane, enhancing adhesion to MHC class II bearing sperm. By interaction with MHC class II on the surface of APC, CD4 initiates the development of T lymphocytes and helps the optimum functioning of mature T lymphocytes. The binding of the GK1.5 antibody blocks the binding of the Anti-Mouse CD4 RM4-5 antibody.

Chromosome 9 consists of about 145 million bases, represents 4% of the human genome and encodes nearly 900 genes. Thought to play a role in gender determination, deletion of the distal portion of 9p can lead to development of male to female sex reversal, the phenotype of a female with a male X,Y genotype. Hereditary hemorrhagic telangiectasia, which is characterized by harmful vascular defects, is associated with the chromosome 9 gene encoding endoglin protein, ENG. Interleukin 12 (IL-12p70) is a heterodimeric cytokine composed of p35 and p40 subunits. IL-12 is produced by monocytes, macrophages, dendritic cells, neutrophils, and B cells in response to bacterial products and cytokines such as IFN-γ. The IL-12 receptor is expressed on T, NK, and dendritic cells. Upon binding, IL-12 initiates signaling via the JAK/STAT signaling pathway and stimulates NK, B, and T cells to produce IFN-γ (Watford <i>et al.</i>). It also regulates cytokine synthesis, proliferation of T and NK cells, and stimulates differentiation of CD4+ and CD8+ T cells (Germann and Rüde). P73 protein is a structural and functional homologue of p53, a tumor suppressor gene. In this study, The p73 protein, p19ras, by the yeast two-hybrid screening method. Alternative splicing of the proto-oncogene H-ras pre-mRNA has led to two distinct transcripts, Ras proteins are known to be small membrane-localized guanine nucleotide-binding proteins. However, unlike other Ras proteins, p19ras is localized in the nucleus and the cytosol and its interaction with P73 protein occurred exclusively in the nucleus. Oncogenic MDM2 (mouse double minutes 2) is a known repressor of p73 transcriptional activity. In this study, when p19ras was bound to MDM2, it further inhibited the association of MDM2 to the p73 protein. Therefore, this study presents a novel pathway of Ras signaling that occurs in the nucleus, involving p19ras and p73.
