PKM, also named as OIP3, PK2, PK3, PK-M1/2, p58, THBP1, CTHBP and Tumor M2-PK, belongs to the pyruvate kinase family. It is glycolytic enzyme that catalyzes the transfer of a phosphoryl group from phosphoenolpyruvate (PEP) to ADP, generating ATP. It stimulates POU5F1-mediated transcriptional activation. PKM plays a general role in caspase independent cell death of tumor cells. The activity of the M2 isoform (but not the M1 isoform) can be inhibited by tyrosine kinase signalling in tumourcells.The primary pyruvate kinase isoform before tumour development is PK-M1; however, the primary isoform from four independent tumours is PK-M2.. HECA, also known as headcase homolog, HDC, HDCL or HHDC, is a 543 amino acid mammalian homolog of the Drosophila headcase protein, a highly basic, cytplasmic peptide that plays a role in mitotic re-entry during adult morphogenesis. Expressed in a variety of tissues with highest expression in thymus, spleen and heart, HECA is thought to play a role in the development of epithelial tube networks in lung tissue and may also be involved in the pathogenesis of lung cancer. The gene encoding HECA maps to human chromosome 6, which contains 170 million base pairs and comprises nearly 6% of the human genome. Deletion of a portion of the q arm of chromosome 6 is associated with early onset intestinal cancer, suggesting the presence of a cancer susceptibility locus. Additionally, Porphyria cutanea tarda, Parkinson's disease, Stickler syndrome and a susceptibility to bipolar disorder are all associated with genes that map to chromosome 6.

P73 protein is a structural and functional homologue of p53, a tumor suppressor gene. In this study, The p73 protein, p19ras, by the yeast two-hybrid screening method. Alternative splicing of the proto-oncogene H-ras pre-mRNA has led to two distinct transcripts, Ras proteins are known to be small membrane-localized guanine nucleotide-binding proteins. However, unlike other Ras proteins, p19ras is localized in the nucleus and the cytosol and its interaction with P73 protein occurred exclusively in the nucleus. Oncogenic MDM2 (mouse double minutes 2) is a known repressor of p73 transcriptional activity. In this study, when p19ras was bound to MDM2, it further inhibited the association of MDM2 to the p73 protein. Therefore, this study presents a novel pathway of Ras signaling that occurs in the nucleus, involving p19ras and p73.

NDE1 (NUDE /NUDE1) is a LIS1-interacting protein. A signaling pathway containing NDE1, LIS1, and cytoplasmic dynein is conserved for eukaryotic nuclear migration and has profound impact on neuronal migration during development of the central nervous system. LIS1 and dynein have been shown to be involved in mitosis and NDE1 participates in a subset of dynein function in M phase. NDE1 is specifically phosphorylated in M phase in its serine/threonine phosphorylation motifs, probably by Cdc2 and also Erk1 and Erk2. A fraction of NDEl binds to centrosomes strongly in interphase and is localized to mitotic spindles in early M phase. Phosphorylation of NDE1 also regulates the cell-cycle-dependent distribution, possibly by increasing its dissociation rate at the microtubule-organizing center.
