A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. Bcl-rambo Antibody: Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. Members of the Bcl-2 family are known to be critical regulators of this process. These proteins are characterized by the presence of several conserved motifs termed Bcl-2 homology (BH) domains. A novel, widely expressed member termed Bcl-rambo was recently identified. This protein is localized to mitochondria in mammalian cells and its overexpression induces apoptosis which could be blocked by co-expression of inhibitor of apoptosis proteins (IAPs) such as XIAP, cIAP1, and cIAP2. Bcl-rambo shows overall homology to the anti-apoptotic members containing BH motifs, but unlike Bcl-2, the C-terminal membrane anchor of Bcl-rambo is preceded by a unique 250 amino acid insertion. This region by itself can induce apoptosis more efficiently than the Bcl-2 homology regions, suggesting that Bcl-rambo may be important other pro-apoptotic pathways.

This antibody recognizes a glycoprotein of 75 kDa, identified as low affinity Nerve Growth Factor (NGF) Receptor (p75NGFR) or Neurotrophin Receptor (p75NTR). Its epitope spans in aa 1-160 of extracellular domain of NGFR/NTR. NGF-receptor contains an extracellular domain containing four 40-amino acid repeats with 6 cysteine residues at conserved positions followed by a serine/threonine-rich region, a single transmembrane domain, and a 155-amino acid cytoplasmic domain. The cysteine-rich region contains the nerve growth factor binding domain. NGF is important for the development, differentiation, and survival of variety of neuronal and non-neuronal cells. Its action is mediated by binding two distinct receptors, the high affinity p140 and low affinity p75.

SATB1 Antibody: Human special AT-rich sequence-binding protein-1 (SATB1) is a nuclear matrix/scaffold-associated region DNA-binding protein, predominantly expressed in the thymus and pre-B cells. Like its homolog SATB2, SATB1 selectively binds double-stranded, special AT-rich DNA sequences in which one strand exclusively consists of well-mixed A, T, and C nucleotides. SATB1 contains a dimerization domain that shares similarity with the PDZ motif, identified as an indispensable element for high-affinity binding of SATB1 to DNA. SATB1 constitutes a functional nuclear architecture that has a 'cage-like' protein distribution surrounding heterochromatin and regulates gene expression through chromatin remodeling/HDAC (histone deacetylase complex) and transcription factors recruitment. SATB1 functions as a 'genome organizer' essential for proper T-cell development. Recent studies show that SATB1 is necessary for breast cancer cells to become metastatic, and when ectopically expressed in non-metastatic cells, can induce invasive activity in vivo. At least two isoforms of SATB1 are known to exist.

EDRF1 (erythroid differentiation-related factor 1), also known as C10orf137 (chromosome 10 open reading frame 137), is a 1,238 amino acid protein containing two TPR repeats. Localizing to nucleus, EDRF1 is involved in transcriptional activation of globin genes by regulating DNA-binding activity of GATA-1 transcription factor. EDRF1 may also play an important role in organ development and histological differentiation. EDRF1 exists as four alternatively spliced isoforms and is encoded by a gene mapping to human chromosome 10q26.13.

DACH1, a homologue of the Drosophila dachshund gene, is a key regulator of cell fate determination during eye, leg, and brain development in the fly. Through interacting with NCoR and Smad4, DACH1 is able to inhibit the transforming growth factor-beta (TGF-beta) signaling pathway. DACH1 can inhibit breast cancer cellular proliferation via cyclin D1, suggesting a possible role in tumor suppression. Additionally, DACH1 plays an important role in negative regulation of RANKL (Receptor activator of NF-kappaB ligand ) gene expression in marrow stromal/preosteoblast cells, and loss of DACH1 expression might be involved in endometrial cancer progression. Four isoforms of DACH1 are produced by alternative splicing, but isoform1(~97kd) is the predominantly expressed form in tissue. This antibody is a mouse monoclonal antibody raised against residues near the C terminus of human DACH1.

The neuronal growth-associated protein GAP43 is also known as neuromodulin, B-50, P-57, F1 and pp46. Deficiency of GAP43 in mice results in death early in the postnatal period. GAP43 is one of the main substrates for protein kinase C in the brain. GAP43 is an intracellular growth-associated protein that appears to assist neuronal pathfinding and branching during development and regeneration, and may contribute to presynaptic membrane changes in the adult, leading to the neurotransmitter release, endocytosis and synaptic vesicle recycling, long-term potentiation, spatial memory formation, and learning. The predicated molecular weight of about 25 kDa is much lower than the apparent observed molecular weight of 43 kDa on SDS-PAGE gels, and this occurs because the highly charged nature of GAP43 causes it to bind less than the average amount of SDS per amino acid, and because the protein has an elongated structure.

Making up nearly 6% of the human genome, chromosome 6 contains around 1,200 genes within 170 million base pairs of sequence. Deletion of a portion of the q arm of chromosome 6 is associated with early onset intestinal cancer, suggesting the presence of a cancer susceptibility locus. Porphyria cutanea tarda is associated with chromosome 6 through the HFE gene which, when mutated, predisposes an individual to developing this porphyria. Notably, the PARK2 gene, which is associated with Parkinson's disease, and the genes encoding the major histocompatiblity complex proteins, which are key molecular components of the immune system and determine predisposition to rheumatic diseases, are also located on chromosome 6. Stickler syndrome, 21-hydroxylase deficiency and maple syrup urine disease are also associated with genes on chromosome 6. A bipolar disorder susceptibility locus has been identified on the q arm of chromosome 6. The C6orf120 gene product has been provisionally designated C6orf120 pending further characterization.
