Attractin is a serum glycoprotein of 175 kDaand found in both membrane-bound and secreted forms as a result of alternative splicing. Both the secreted and membrane-bound forms of attractin may be involved in the development and maintenance of the central nervous system. Membrane-bound attractin is a co-receptor for Agouti, antagonist of melanocortin-1 receptor. Secreted attractin, expressed by activated T lymphocytes and modulates interactions between T cells and monocytes/macrophages, was examined as a potential marker of immune activity. Attractinmay be a component of a pathway for regulated protein turnover that also involves mahogunin, a wide-expressed E3 ubiquitinligase found at particularly high levels in the brain. Nudel is important for normal cortical development. It is invovled in microtubule organization, nuclear translocation, and neuronal positioning in concert with various other factors (including Lis1, Pafah1b1, Pahfah1b2, dynein, dynorphin A and cdk5). Western blot analysis of mouse tissues shows abundant expression of Nudel in brain and testis, and much lower expression in heart, liver, kidney, and skeletal muscle. In fractionated rat brain, Nudel and Lis1 are both found in fractions enriched for postsynaptic density proteins. Immunostaining of embryonic day 18 mouse brain sections revealed staining of migrating neurons and thalamocortical axons of the intermediate zone of the developing cerebral cortex, as well as several other developing brain regions. The deduced protein contains 345 amino acids and has a calculated molecular mass of 38.4 kDa. It has a coiled coil motif (residues 19 to 201), followed by several potential phosphorylation sites for casein kinase II, protein kinase C or CDK5. Nudel shares about 50% identity with mouse and human NUDE proteins.

The FOX family of transcription factors share a common DIUA binding domain termed a winged-helix or forkhead domain. Many FOX proteins play important roles in development, metabolism, cancer and aging. FOXD1 (also designated Brain Factor 2 or BF-2) is involved in regulating inflammation as well as kidney and retinal development. FOXD1 regulates the activity of NF-AT and NFkB. Deficiency of FOXD1 results in multiorgan, systemic inflammation, exaggerated Th cell-derived cytokine production, and T cell proliferation in autogolgous MLRs. In kidneys, FOXD1 controls the production of signals required for the normal transition of induced mesenchyme into tubular epithelium and full growth and branching of the collecting system. Deletion of FOXD1 results in renal abnormalities. FOXD2 acts as a modulator of T cell activation.

The glutamate receptor interacting proteins, GRIP1 and GRIP2, members of the PDZ domain-containing protein family, mediate the trafficking and membrane organization of a number of transmembrane proteins. GRIP1 and GRIP2 specifically bind to the AMPA receptor subunits, GluR 2/3 and are involved in the targeting of GluR 2/3 to the synapse. GRIP1 is expressed in early development before the expression of AMPA receptors, while GRIP2 expression parallels that of AMPA receptors during later developmental stages. GRIP1 and GRIP2 may be involved in the induction of cerebellar long-term depression (LTD).
