Egr2 belongs to the Egr family that binds to DNA through highly conserved zinc finger domains and has a pivotal role in hindbrain development and myelination of the peripheral nervous system, development of T and/or B cells . It has been demonstrated that Egr2 is induced early upon TCR engagement in a NFAT-dependent manner, and that overexpression of Egr2 inhibits T cell activation. Overexpressing Egr2 shows an upregulation of E3-ligase Cbl-band reduced production of interleukin-2 (IL-2), suggesting that Egr2 is important for the maintenance of T cell tolerance by negatively regulating T cell activation.

It plays a role in both erythroid cell and osteoblast differentiation during development, inhibiting the activities of early B-cell factor 1 (EBF1) in erythroid cells and Runx2 in osteoblast precursors. The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth, and invasion. This gene is located in a region on chromosome X, which is associated with Borjeson-Forssman-Lehmann syndrome (BFLS), making it a possible candidate gene for familial cases of the BFLS, and for other syndromal and nonspecific forms of X-linked mental retardation mapping to this region. Fibroblast growth factor 13 (FGF13) is probably involved in nervous system development and function. Alternative splicing of this gene results in several transcript variants encoding different isoforms with different N-termini.
