Egr2 belongs to the Egr family that binds to DNA through highly conserved zinc finger domains and has a pivotal role in hindbrain development and myelination of the peripheral nervous system, development of T and/or B cells . It has been demonstrated that Egr2 is induced early upon TCR engagement in a NFAT-dependent manner, and that overexpression of Egr2 inhibits T cell activation. Overexpressing Egr2 shows an upregulation of E3-ligase Cbl-band reduced production of interleukin-2 (IL-2), suggesting that Egr2 is important for the maintenance of T cell tolerance by negatively regulating T cell activation.

The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth, and invasion. This gene is located in a region on chromosome X, which is associated with Borjeson-Forssman-Lehmann syndrome (BFLS), making it a possible candidate gene for familial cases of the BFLS, and for other syndromal and nonspecific forms of X-linked mental retardation mapping to this region. Fibroblast growth factor 13 (FGF13) is probably involved in nervous system development and function.
