Docking protein 1 is constitutively tyrosine phosphorylated in hematopoietic progenitors isolated from chronic myelogenous leukemia (CML) patients in the chronic phase. It may be a critical substrate for p210(bcr/abl), a chimeric protein whose presence is associated with CML. Docking protein 1 contains a putative pleckstrin homology domain at the amino terminus and ten PXXP SH3 recognition motifs. Docking protein 2 binds p120 (RasGAP) from CML cells. It has been postulated to play a role in mitogenic signaling.

May be a physiological regulator of [1-14C]-2-deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblasts. CD80 is a member of the Ig superfamily and serves as the ligand for two T cell molecules, CD28 and CTLA4. Interactions between CD28 and CD80 on activated B cells result in enhanced T cell activation. CD80 is rapidly induced on the surface of in vitro activated B cells, Epstein Barr Virus (EBV) transformed B cell lines, Burkitts lymphoma cell lines, freshly isolated follicular B lymphoma cells, T cells, and monocytes. It is also expressed at high levels in dendritic cells. It reacts weakly with a small proportion of non activated normal B cells and with HTLV1 infected T cells. CD80 does not react with peripheral monocytes, resting and activated normal T cells, T cell lines and T cell clones, nor with myelomonocytic cell lines.

CD97, a CD55 Receptor, is a leukocyte antigen located on most activated leukocytes. CD97 is highly homologous to EMR2 with five EGF-like domains in the N-terminal extracellular domain, suggesting that it may be involved in cell signaling as well as in cell adhesion (Hamann et al., 1996). Two variants are produced by alternative splicing, where the shorter form lacks exons 5 and 6 and reduces the number of EGF-like domains from 5 to 3, compared to the full-length isoform. CD97 is associated with with the activated lesions of multiple sclerosis. CD97 expression is also implicated in the dedifferentiation of colon and thyroid tumors. CD97 is expressed abundantly in cells of hematopoietic origin and is markedly upregulated on activated T and B cells. ESTs have been isolated from a wide variety of tissue libraries.

SH3BP4 Antibody: SH3BP4 was initially isolated as a novel cDNA using differential display analysis using cultured human corneal fibroblasts. The protein contains three Asn-Pro-Phe (NPF) motifs, an SH3 domain, a PXXP motif, a bipartite nuclear targeting signal, and a tyrosine phosphorylation site. SH3BP4 has been shown to interact with specific endocytic proteins such as clathrin, dynamin, and the transferrin receptor (TfR) and localizes to TfR-containing coated pits and vesicles. It is thought to be involved in cargo-specific control of clathrin-mediated endocytosis, specifically controlling the internalization of TfR. At least two isoforms of SH3BP4 are known to exist.

The calcium-sensing receptor protein (CASR) is a G protein-coupled receptor that is expressed in the parathyroid hormone (PTH)-producing chief cells of the parathyroid gland, and the cells lining the kidney tubule. It senses small changes in circulating calcium concentration and couples this information to intracellular signaling pathways that modify PTH secretion or renal cation handling, thus this protein plays an essential role in maintaining mineral ion homeostasis. Mutations in this gene cause familial hypocalciuric hypercalcemia, familial, isolated hypoparathyroidism, and neonatal severe primary hyperparathyroidism. Recent evidence suggests that activated CASR contributes to the cytokine secretion through the partial MAPK and NF-kappaB pathways in T cells.

CD45, also known as leukocyte common antigen (LCA), B220, and T200, is a 180-240 kD single chain type I membrane glycoprotein. It is a tyrosine phosphatase expressed on the plasma membrane of all hematopoietic cells, except erythrocytes and platelets. CD45 is a signaling molecule that regulates a variety of cellular processes including cell growth, differentiation, cell cycle, and oncogenic transformation. CD45 is one of the best negative selection markers for characterizing and/or isolating human mesenchymal stem cells from bone marrow and other sources. Along with the positive selection markers (such as CD29, CD44, CD90, CD105 and CD166), negative selection markers (such as CD34 and CD45) are used for MSC identification.

TNIK is a MSN protein kinase that interacts with both TNF receptor-associated factor 2 (TRAF2) and the adapter protein NCK. The protein has been shown to activate the c-Jun N-terminal kinase pathway when over expressed in Phoenix-A cells. TNIK has been shown to phosphorylate gelsolin, the principal intracellular and extracellular actin-severing protein, in vitro. This and evidence from mutational studies suggest that TNIK functions in the regulation of the cytoskeleton. Northern analysis indicates TNIK expression in human heart, skeletal muscle, and brain, with lower levels of expression in kidney, liver, lung, and pancreas. ESTs have been isolated from human tissue libraries, including normal amnion, gallbladder and skin.
