The gene product is a member of the cadherin superfamily, a group of integral membrane proteins characterized by the presence of cadherin-type repeats. In addition to containing 34 tandem cadherin-type repeats, the gene product has five epidermal growth factor (EGF)-like repeats and one laminin A-G domain. This gene is expressed at high levels in a number of fetal epithelia. Its product probably functions as an adhesion molecule and/or signaling receptor, and is likely to be important in developmental processes and cell communication. CD40 Ligand (CD40LG) is a type II transmembrane glycoprotein that belongs to the TNF superfamily. Like other TNF superfamily members, CD40LG exists as a trimer in membrane bound and soluble form, both of which are bioactive. CD40LG is a ligand for CD40; its ligation also initiates signal transduction in CD40LG expressing cells. CD40LG is a differentiation antigen that is expressed on the surface of T-cells. It stimulates B-cell proliferation and secretion of all immunoglobulin isotypes in the presence of cytokines. CD40LG has been shown to induce cytokine production and tumoricidal activity in peripheral blood monocytes. It also co-stimulates proliferation of activated T-cells and this is accompanied by the production of IFN-gamma, TNF-alpha, and IL2.

Familial dysautonomia is also associated with chromosome 9 though through the gene IKBKAP. Notably, chromosome 9 encompasses the largest interferon family gene cluster. Chromosome 9 is partnered with chromosome 22 in the translocation leading to the aberrant production of BCR-ABL fusion protein often found in leukemias. The C9orf72 gene product has been provisionally designated C9orf72 pending further characterization. There are two isoforms of C9orf72 that are produced as a result of alternative splicing events.
