SCO2 Antibody: Synthesis of cytochrome c oxidase 2 was initially identified in yeast as one of two cytochrome c oxidase (COX) assembly proteins that enable the assembly of cytochrome c holoenzyme, a complex that catalyzes the transfer of reducing equivalents from cytochrome c to molecular oxygen and pumps protons across the inner mitochondrial membrane. Like their yeast homologs, the function of both SCO2 and SCO1 are dependent on copper ion binding. Recent studies suggest that SCO2 expression is regulated by p53, so that a decrease in p53 expression, such as in numerous tumors and cells lines, the drop in SCO2 expression leads to a shift from normal aerobic respiration towards the production of glycolytic ATP. Defects in the SCO2 protein are also associated with fatal infantile cardioencephalomyopathy and COX deficiency.

The protein encoded by this gene is part of a complex of proteins that constitute adherens junctions (AJs). AJs are necessary for the creation and maintenance of epithelial cell layers by regulating cell growth and adhesion between cells. The encoded protein also anchors the actin cytoskeleton and may be responsible for transmitting the contact inhibition signal that causes cells to stop dividing once the epithelial sheet is complete. Finally, this protein binds to the product of the APC gene, which is mutated in adenomatous polyposis of the colon. Mutations in this gene are a cause of colorectal cancer (CRC), pilomatrixoma (PTR), medulloblastoma (MDB), and ovarian cancer. Cyclin O Antibody: Cyclin O, also known as CCNO, has recently been identified as a Cdk1- and Cdk2-activating cyclin specific to apoptosis in lymphoid cells. Cyclin O binds to and activates Cdk2 in response to instrinsic apoptotic stimuli such as glucocorticoids or DNA-damaging agents. Supression of Cyclin O expression by shRNA leads to the inhibition of glucocorticoid and DNA-damage-induced apoptosis due to a failure of apical caspase activation while leaving the CD95 death receptor-mediated apoptosis intact. Note: this gene, which had a previous symbol of UNG2, was erroneously identified as a uracil DNA glycosylase. A later publication identified this gene's product as a cyclin protein family member.

The 15.2 antibody reacts with the 85-110 kDa intracellular adhesion molecule-1 (ICAM-1), a member of the Ig superfamily which acts as a ligand for the Lymphocyte Function-Associated Antigen-1 (LFA-1). TIM-4 Antibody: The T cell immunoglobulin and mucin domain containing protein (TIM) family encodes cell surface receptors that are involved in the regulation of T helper (Th) -1 and -2 cell-mediated immunity. TIM-4, which is preferentially expressed on macrophages and dendritic cells, is the natural ligand of TIM-1, and this binding leads to T-cell expansion and cytokine production. Unlike other members of the TIM family, TIM-4 lacks a putative tyrosine phosphorylation signal sequence in its intracellular domain. The PVM13-1 monoclonal antibody bings to human IL-13. IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and also by mast cells and NK cells. Targeted deletion of IL-13 in mice resulted in impaired Th2 cell development and indicated an important role for IL-13 in the expulsion of gastrointestinal parasites. IL-13 exerts anti-inflammatory effects on monocytes and macrophages and it inhibits the expression of inflammatory cytokines such as IL-1β, TNF-α, IL-6 and IL-8. IL-13 has also been shown to enhance B cell proliferation and to induce isotype switching, resulting in increased production of IgE. Blocking of IL-13 activity inhibits the pathophysiology of asthma. The PVM13-1 antibody is reported to be a neutralizing antibody.

This protein is a component of anaphase-promoting complex (APC), which is composed of eight protein subunits and highly conserved in eucaryotic cells. APC catalyzes the formation of cyclin B-ubiquitin conjugate that is responsible for the ubiquitin-mediated proteolysis of B-type cyclins. This protein and 3 other members of the APC complex contain the TPR (tetratricopeptide repeat), a protein domain important for protein-protein interaction. This protein was shown to interact with mitotic checkpoint proteins including Mad2, p55CDC and BUBR1, and thus may be involved in controlling the timing of mitosis. NF-kappa B mediates the expression of a great variety of genes in response to extracellular stimuli including IL-1, TNF alpha , LPS and mitogens. A serine/threonine protein kinase which mediates NF-kappa B activation by IL-1, TNF alpha and CD95 was identified recently and designated NIK (for NF-kappa B inducing kinase). NIK is an activator of I kappa B kinase alpha and beta (IKK alpha and IKK beta ). Therefore, NIK is a key molecule in the NF-kappa B signaling pathway leading to the induction of a variety of gene expression in response to proinflammatory cytokines and bacteria products.

PION Antibody: Accumulation of the amyloid-beta peptide (Abeta) in the cerebral cortex is a critical event in the pathogenesis of Alzheimer's disease. The beta-amyloid protein precursor (APP) is cleaved by one of two beta-secretases (BACE and BACE2), producing a soluble derivative of the protein and a membrane anchored 99 -amino acid carboxy-terminal fragment (C99). The C99 fragment serves as substrate for gamma-secretase to generate the 4 kDa amyloid-beta peptide (Abeta), which is deposited in the Alzheimer's disease patient's brains. PION, or GSAP, selectively increases amyloid-beta production through a mechanism involving its interaction with both gamma-secretase and the APP C-terminal fragment, suggesting that PION may be a potential therapeutic target for the treatment of Alzheimer's disease.
