Description:
The pleiotropic IL3 (Interleukin 3) is a 15kDa cytokine that is primarily secreted by activated T lymphocytes and stimulates the proliferation and differentiation of hematopoietic cells. IL3 acts in hematopoiesis by controlling the production, differentiation, and function of two related white cell populations of the blood, the granulocytes and the monocytes/macrophages. It induces granulocytes, macrophages, mast cells, stem cells, erythroid cells, eosinophils and megakaryocytes and is expressed by activated T cells, mast cells, and natural killer cells. IL3 not only supports growth of both pluripotent stem cells and the more differentiated committed progenitors, but it also stimulates the functional activity of some fully differentiated cells. IL3 has also been shown to protect mast cells from undergoing apoptosis.

Description:
The PVM13-1 monoclonal antibody bings to human IL-13. IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and also by mast cells and NK cells. Targeted deletion of IL-13 in mice resulted in impaired Th2 cell development and indicated an important role for IL-13 in the expulsion of gastrointestinal parasites. IL-13 exerts anti-inflammatory effects on monocytes and macrophages and it inhibits the expression of inflammatory cytokines such as IL-1β, TNF-α, IL-6 and IL-8. IL-13 has also been shown to enhance B cell proliferation and to induce isotype switching, resulting in increased production of IgE. Blocking of IL-13 activity inhibits the pathophysiology of asthma. The PVM13-1 antibody is reported to be a neutralizing antibody.

The protein encoded by this gene shares strong similarity with Saccharomyces cerevisiae protein Cdc27, and the gene product of Schizosaccharomyces pombe nuc 2. This protein is a component of anaphase-promoting complex (APC), which is composed of eight protein subunits and highly conserved in eucaryotic cells. APC catalyzes the formation of cyclin B-ubiquitin conjugate that is responsible for the ubiquitin-mediated proteolysis of B-type cyclins. This protein and 3 other members of the APC complex contain the TPR (tetratricopeptide repeat), a protein domain important for protein-protein interaction. This protein was shown to interact with mitotic checkpoint proteins including Mad2, p55CDC and BUBR1, and thus may be involved in controlling the timing of mitosis. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.

AIF Antibody: Apoptosis is characterized by several morphological nuclear changes including chromatin condensation and nuclear fragmentation. These changes are triggered by the activation of members of caspase family, caspase activated DNase, and several novel proteins. A novel gene, the product of which causes chromatin condensation and DNA fragmentation, was recently identified, cloned, and designated apoptosis inducing factor (AIF). Like the critical molecules, cytochrome c and caspase-9, in apoptosis, AIF localizes in mitochondria. AIF translocates to the nucleus when apoptosis is induced and induces mitochondria to release the apoptogenic proteins cytochrome c and caspase-9. AIF induces chromatin condensation and DNA fragmentation, which are the hallmarks of apoptosis, of the isolated nucleus and the nucleus in live cells by microinjection. AIF is highly conserved between human and mouse and widely expressed.

Similarly, the lipopolysaccharide of Neisseria meningitidis is highly toxic, as it has an additional virulence factor in the form of its antiphagocytic capsule. Both pathogens produce IgA proteases which promote virulence. Many normal individuals may harbor Neisseria meningitidis in the upper respiratory tract, but Neisseria gonorrhoeae is never part of the normal flora and is only found after sexual contact with an infected person (or direct contact, in the case of infections in the newborn).

Cochlin is an extracellular matrix protein expressed in the cochlea and vestibule of the inner ear. Mutations in the cochlin gene are linked to the autosomal-dominant DFNA9 syndrome, a disorder characterized by hearing loss. Cochlin is also expressed in follicular dendritic cells of the spleen and the lymph nodes. The N-terminus of cochlin contains a LCCL domain also present in Factor C, a serine protease of the horseshoe crab Limulus, involved in activating the coagulation cascade in response to LPS. A recent study showed that in response to inflammation, LPS or bacterial infection, cochlin is cleaved by the proteases aggrecanases and secreted into blood circulation. The secreted cochlin (LCCL domain) seems to have a role in local cytokine production, recruitment of immune effector cells and bacterial clearance. Cochlin is secreted as two forms p8 and p18, whereas p18 being the glycosylated form of p8.
