Description:
M-GM-CSF is produced in response to a number of inflammatory mediators by mesenchymal cells present in the hemopoietic environment and at peripheral sites of inflammation. GM-CSF is able to stimulate the production of neutrophilic granulocytes, macrophages, and mixed granulocyte-macrophage colonies from bone marrow cells and can stimulate the formation of eosinophil colonies from fetal liver progenitor cells. GM-CSF can also stimulate some functional activities in mature granulocytes and macrophages. GM-CSF receptors shows significant homologies with other receptors for hematopoietic growth factors, including IL2 -beta, IL-3, IL-6, IL-7, EPO and the Prolactin receptors. Recombinant mouse GM- CSF produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 125 amino acids and having a molecular mass of 14,285.35 Daltons.

Alzheimer's disease (AD) patients with an inherited form of the disease carry mutations in the presenilin proteins (PSEN1; PSEN2) or the amyloid precursor protein (APP). These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that they either directly regulate gamma-secretase activity or themselves are protease enzymes. Two alternative transcripts of PSEN2 have been identified.

ANKRD54 is a 300 amino acid protein that contains four ankyrin repeats and is expressed as two alternatively spliced isoforms. The gene encoding human ANKRD54 maps to chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, neurofibromatosis type 2, autism and schizophrenia. Additionally, translocations between chromosomes 9 and 22 may lead to the formation of the Philadelphia chromosome and the subsequent production of the novel fusion protein BCR-Abl, a potent cell proliferation activator found in several types of leukemias.

IL-36alpha (IL-1F6), IL-36beta (IL-1F8) and IL-36gamma (IL-1F9) bind to IL-36R (IL-1Rrp2) and IL-1RAcP, activating similar intracellular signals as IL-1. IL-36Ra inhibits the production of proinflammatory cytokines, including IL-12, IL-1beta, IL-6, TNF-alpha and IL-23 induced by IL-36 in BMDC and CD4 T cells. Skin and dendritic cells are targets of the IL-36 interleukins leading to a Th1 response. These cytokines may represent potential targets for immune-mediated inflammatory conditions or, alternatively, could be used as adjuvants in vaccination.

The 15.2 antibody reacts with the 85-110 kDa intracellular adhesion molecule-1 (ICAM-1), a member of the Ig superfamily which acts as a ligand for the Lymphocyte Function-Associated Antigen-1 (LFA-1). ICAM-1 is also known CD 54 and is expressed on non-hematopoietic cells of vascular endothelial, thymic epithelial, fibroblasts lineages, and on hematopoietic cells like macrophages, mitogen-stimulated T-lymphoblasts, dendritic cells in tonsils, lymph nodes and Peyer’s patches, and germinal center B cells.Inflammatory mediators (IL-1, TNF, IFN-γ) enhance the production of ICAM-1 on fibroblasts and endothelial cells within few hours. This gene encodes a member of the Wiskott-Aldrich syndrome protein family. The gene product is a protein that forms a multiprotein complex that links receptor kinases and actin. Binding to actin occurs through a C-terminal verprolin homology domain in all family members. The multiprotein complex serves to tranduce signals that involve changes in cell shape, motility or function. The published map location (PMID:10381382) has been changed based on recent genomic sequence comparisons, which indicate that the expressed gene is located on chromosome 1, and a pseudogene may be located on chromosome X. Two transcript variants encoding different isoforms have been found for this gene.

Cyclin O Antibody: Cyclin O, also known as CCNO, has recently been identified as a Cdk1- and Cdk2-activating cyclin specific to apoptosis in lymphoid cells. Cyclin O binds to and activates Cdk2 in response to instrinsic apoptotic stimuli such as glucocorticoids or DNA-damaging agents. Supression of Cyclin O expression by shRNA leads to the inhibition of glucocorticoid and DNA-damage-induced apoptosis due to a failure of apical caspase activation while leaving the CD95 death receptor-mediated apoptosis intact. Note: this gene, which had a previous symbol of UNG2, was erroneously identified as a uracil DNA glycosylase. A later publication identified this gene's product as a cyclin protein family member.
