PARP-1 (ARTD1) is involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks. PARP-1 positively regulates the transcription of MTUS1 and negatively regulates the transcription of MTUS2/TIP150. It forms a complex with EEF1A1 and TXK that acts as a T-helper 1 (Th1) cell-specific transcription factor and binds the promoter of IFN-gamma to directly regulate its transcription, and is thus involved importantly in Th1 cytokine production. PARP-1 (E998K mutant) is an inactive form of PARP-1 which can be used as a control compound.

Folliculin is thought to act as a tumor suppressor as mutations or loss of heterozygosity in this gene are associated with BHD syndrome-related renal tumors. Folliculin and FNIP1, a protein that shares 49% identity to FNIP2, bind to AMPK, an important energy sensor in cells that negatively regulates the mammalian target of rapamycin (mTOR), a protein that is thought to be the master switch for cell growth and proliferation. FNIP1 and FNIP2 are able to form homo- and heteromeric multimers, suggesting these proteins may have a functional relationship.

APH1 Antibody: APH1 was initially identified as a component of the Notch pathway in C. elegans. Along with nicastrin, PEN2, and presenilin-1 APH1 is an essential component of the gamma-secretase complex which cleave the amyloid precursor protein (APP) at what are known as the gamma- and epsilon-sites and can lead to the accumulation of the Amyloid beta peptide (Abeta) cleavage product that is associated with Alzheimer's disease. APH1 exists in at least three distinct isoforms with APH1a as the principal isoform present in the gamma-secretase complex. Mice deficient in this isoform, but not the other two, were lethal at E10.5, with impaired vascular and neural development observed.

These proteins all have similar functions as they all mediate extracellular matrix organization and act as binding partners of TGF beta. Osteoglycin, which also may be designated osteoinductive factor (OIF), is a secreted protein detected in bone tissues. Osteoglycin induces the formation of bone in conjunction with either TGF-beta-1 or TGF-beta-2. The precursor form of the OGN gene product, designated Mimecan, is subject to in situ proteolytic cleavage to yield the mature Osteoglycin.

The ALDH1L2 gene has three transcriptional variants with the molecular weight of 102 kDa, 42 kDa and 89 kDa. It is a mitochondrial enzyme, a likely source of CO2 production from 10-formyltetrahydrofolate in mitochondria and playing an essential role in the distribution of one-carbon groups between the cytosolic and mitochondrial compartments of the cell. It also has dehydrogenase/hydrolase activities similar to cytosolic FDH (ALDH1L1). The native ALDH1L2 has been reported to exist as a dimer or tetramer.

Finally, this protein binds to the product of the APC gene, which is mutated in adenomatous polyposis of the colon. Mutations in this gene are a cause of colorectal cancer (CRC), pilomatrixoma (PTR), medulloblastoma (MDB), and ovarian cancer.
