Mouse interleukin 13 (mIL-13) is a pleiotropic cytokine produced by activated Th2 cells. IL-13 induces B cell proliferation and immunoglobin production. It contains a four helical bundle with two internal disulfide bonds. Mouse IL13 shares 58% sequence identity with human protein and exhibits cross-species activity. IL13 signals via receptor IL13R (type2, IL4R) and activates STAT-6. IL13 initially binds IL-13R alpha1 with low affinity and triggers association of IL4R alpha, generating a high affinity heterodimeric receptor IL13R and eliciting downstream signals. IL13 also binds IL-13R alpha2 with high affinity, which plays a role in a negative feedback system of IL13 signaling. IL13 is an important mediator of allergic inflammation and disease.

Cochlin is an extracellular matrix protein expressed in the cochlea and vestibule of the inner ear. Mutations in the cochlin gene are linked to the autosomal-dominant DFNA9 syndrome, a disorder characterized by hearing loss. Cochlin is also expressed in follicular dendritic cells of the spleen and the lymph nodes. The N-terminus of cochlin contains a LCCL domain also present in Factor C, a serine protease of the horseshoe crab Limulus, involved in activating the coagulation cascade in response to LPS. A recent study showed that in response to inflammation, LPS or bacterial infection, cochlin is cleaved by the proteases aggrecanases and secreted into blood circulation. The secreted cochlin (LCCL domain) seems to have a role in local cytokine production, recruitment of immune effector cells and bacterial clearance. Cochlin is secreted as two forms p8 and p18, whereas p18 being the glycosylated form of p8.

This protein forms a trimeric complex with cyclin H and MAT1, which functions as a Cdk-activating kinase (CAK). Translocation of portions of chromosome 8 with amplifications of the c-Myc gene are found in some leukemias and lymphomas, and typically associated with a poor prognosis. Portions of chromosome 8 have been linked to schizophrenia and bipolar disorder. Trisomy 8, also known as Warkany syndrome 2, most often results in early miscarriage but is occasionally seen in a mosaic form in surviving patients who suffer to a varying degree from a number of symptoms including retarded mental and motor development, and certain facial and developmental defects. WRN is a DNA helicase encoded by chromosome 8 and shown defective in those with the early aging disorder Werner syndrome. Chromosome 8 is also associated with Pfeiffer syndrome, congenital hypothyroidism and Waardenburg syndrome. The C8orf31 gene product has been provisionally designated C8orf31 pending further characterization.

Thought to play a role in gender determination, deletion of the distal portion of 9p can lead to development of male to female sex reversal, the phenotype of a female with a male X,Y genotype. Hereditary hemorrhagic telangiectasia, which is characterized by harmful vascular defects, is associated with the chromosome 9 gene encoding endoglin protein, ENG. Familial dysautonomia is also associated with chromosome 9 though through the gene IKBKAP. Notably, chromosome 9 encompasses the largest interferon family gene cluster. Chromosome 9 is partnered with chromosome 22 in the translocation leading to the aberrant production of BCR-ABL fusion protein often found in leukemias.

The conversion of arachidonic acid to prostaglandin H2 (PGH2) by cyclooxygenase (COX) enzymes is the first step in the synthesis of prostanoids. Prostaglandin D2 (PGD2) is synthesized from PGH2 by the action of PGD synthase, and its actions in vasodilation and platelet activation are mediated via activation of two identified G-protein coupled receptors (GPCRs): DP1 and DP2 (formerly CRTH2). Both DP1 and DP2 receptors are coupled to Gs alpha subunits, resulting in increased adenylyl cyclase activity and cAMP production upon agonist binding. DP1 receptor is expressed moderately in the ileum and weakly in the lung, stomach, and uterus of mice, and weakly in the small intestine of humans. In rodents, expression of PGD synthase and the DP1 receptor is seen in the leptomeninges, where it is believed they play a role in the induction of sleep. The DP1 receptor may also oppose DP2-mediated activation of basophils, eosinophils, and Th2 cells.
