The Bloom syndrome gene product is a member of the RecQ helicase protein family. The protein contains 1417 amino acids; including one ATP binding site, one DEAH box, and two putative nuclear localization signals, as well as phosphorylation sites. BLS protein is known to associate and be phosphorylated by the ATR (ataxia telangiectasia, rad3+) protein, in response to genetic stress. The Bloom's syndrome gene product is thought to play a role in the etiology of two major human health problems, each of which is a very common complication of Bloom's syndrome, namely cancer and diabetes.

The gene encoding C20orf24 maps to human chromosome 20, which houses over 600 genes, some of which are associated with Creutzfeldt-Jakob disease, amyotrophic lateral sclerosis, spinal muscular atrophy, ring chromosome 20 epilepsy syndrome and Alagille syndrome. Additionally, chromosome 20 contains a region with numerous genes which are thought important for seminal production and may be potential targets for male contraception. The PRNP gene encoding the prion protein associated with spongiform encephalopathies, like Creutzfeldt-Jakob disease, is found on chromosome 20. The C20orf24 gene product, also desginated Rab5-interacting protein (RIP5), may be involved in the induction of both caspase-dependent apoptosis and caspase-independent cell death.

In spite of the structural and functional differences, the CDK inhibitor isoforms and the ARF product encoded by this gene, through the regulatory roles of CDK4 and p53 in cell cycle G1 progression, share a common functionality in cell cycle G1 control.
