Interleukin-4 (IL-4) is a cytokine produced by type 2 helper T cells, the Th2 cells. These cells tends to make a specific set of lymphokines including IL-4, IL-5, IL-6, IL-10, IL-13, IL-3 and GM-CSF and fail to produce IL-2, IFN-gamma, and lymphotoxin (TNF-beta). In addition, mast cells can produce IL-4. IL-4 exerts numerous effects on various hematopoietic cell types. On B cells, IL-4 promotes immunoglobulin class switching to IgE and IgG1 isotypes and upregulates MHC class II and CD23 expression. IL-4 promotes survival, growth, and differentiation of both T and B lymphocytes, mast cells and endothelial cells. In addition, IL-4 inhibits the production of TNF, IL-1, and IL-6 by macrophages.

The lymphocyte activation gene-3 (LAG-3, CD223), a member of the immunoglobulin superfamily (IgSF) related to CD4, binds to the major histocompatibility complex (MHC) class II molecules but with higher affinity than CD4. Several alternative mRNA splice-variants of human LAG-3 have been described, two of them encoding potential secreted forms: LAG-3V1 (i.e. the D1-D2 domains of the protein, 36 kDa) and LAG-3V3 (D1-D3, 52 kDa). The longer form was detected by ELISA in the serum of healthy individuals as well as of tuberculosis patients with a favorable outcome. Unmethylated CG dinucleotides within particular sequence contexts are responsible for the immunostimulatory activity of bacterial DNA. Synthetic oligonucleotides (ODN) that contain such CpG motifs (CpG ODNs) mimic microbial DNA. The innate immune system of vertebrates has the ability to recognize CpG motifs in microbial DNA via the Toll-like receptor (TLR) 9 if the CpG ODN were free of additional immune stimulatory contaminants often present in synthetic commercial CpG ODN preparations designed for molecular biology applications (i.e. PCR). Given that high quality CpG ODNs were used, a close link has been established between the expression of TLR9 on certain immune cell subsets and the modulation of the immune system by CpG DNA. Different types of CpG ODNs were identified based on their differing biological effects on different cell types: ODN Type A is a potent inducer of IFN-alpha in human PDC, (i.e. ODN 1585 or 2216) leading to antigen presenting cell (APC) maturation, whereas ODN Type B (i.e. ODN 2006 or ODN 1668 / ODN 1826) is a weak inducer of IFN-alpha but rather stimulates IL-8 production and increasing costimulatory and Ag-presenting molecules and triggers proliferation of B-cells and IgM and IL-6 production. The NUDCD1 gene encodes the 583 amino acid CML66 protein which demonstrates expression in a variety of solid tumor cell lines and leukemias. Expression of CML66 in normal tissues is restricted to testis and heart. The CML66 antibody is found in sera of 18-38% of patients with lung cancer, melanoma or prostate cancer. These findings indicate that CML66 may be immunogenic in a wide variety of malignancies and may be a target for antigen-specific immunotherapy. A short isoform of CML66, designated CML66-S, is a product of alternative splicing. CML66-S has the same C-terminus as full-lenghth CML66 (also designated CML66-L), but has a unique N-terminus. CML66-S shows predominant expression in testis, and is also expressed in very low levels in tumor cells, while CML66 is expressed in tumor cells and testis.
