Description:
T cells require a signal induced by the engagement of the T cell receptor and a costimulatory signal(s) through distinct T cell surface molecules for optimal T cell activation and tolerance. CD273 (PD-L2) is one of two ligands for programmed death-1 (PD-1; CD279), a member of the CD28 family of immunoreceptors. The other identified ligand is PD-L1. CD273 is broadly expressed and also up regulated in a variety of tumor cell lines. On previously activated T cells, CD273 interaction with PD-1 inhibits TCR mediated proliferation and cytokine production, suggesting an inhibitory role in regulating immune responses. CD273 has a costimulatory function on resting T cells activated with suboptimal TCR signals.

They are abundant in the granules of neutrophils and also found in the epithelia of mucosal surfaces such as those of the intestine, respiratory tract, urinary tract, and vagina. Members of the defensin family are highly similar in protein sequence and distinguished by a conserved cysteine motif. Several alpha defensin genes are clustered on chromosome 8. This gene differs from other genes of this family by an extra 83-base segment that is apparently the result of a recent duplication within the coding region. The protein encoded by this gene, defensin, alpha 4, is found in the neutrophils; it exhibits corticostatic activity and inhibits corticotropin stimulated corticosterone production.

Binding to actin occurs through a C-terminal verprolin homology domain in all family members. The multiprotein complex serves to tranduce signals that involve changes in cell shape, motility or function. The published map location (PMID:10381382) has been changed based on recent genomic sequence comparisons, which indicate that the expressed gene is located on chromosome 1, and a pseudogene may be located on chromosome X. Two transcript variants encoding different isoforms have been found for this gene.

Functional analysis has demonstrated that this protein stimulates the dissociation of GDP from RAS protein. The studies of the similar gene in mouse suggested that the Ras-GEF activity of this protein in brain can be activated by Ca2+ influx, muscarinic receptors, and G protein beta-gamma subunit. Mouse studies also indicated that the Ras-GEF signaling pathway mediated by this protein may be important for long-term memory.
