Adiponectin is an adipose-derived secreted protein containing 226 amino acid residues. It is relatively abundant in humans and rodents, accounting for about 0.01% of total plasma protein. The circulating levels of adiponectin are decreased under conditions of obesity, insulin resistance, and type II diabetes. Disruption of adiponectin in mice causes insulin resistance and neointimal formation. Conversely, administration of recombinant adiponectin suppresses hepatic glucose production, and reverses insulin resistance associated with both lipoatrophy and obesity. The protective role of adiponectin is attributed to its anti-inflammatory properties (e.g. ability to suppress expression of TNF-α and class A scavenger receptor in macrophages). Recombinant adiponectin is a multimeric glycoprotein containing amino acids Glu-19 to Asn-244 of the adiponectin precursor protein fused to an N-terminal histidine tag. Monomeric glycosylated adiponectin migrates at an apparent molecular weight of approximately 35.0 kDa by SDS PAGE analysis under reducing conditions.

IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and also by mast cells and NK cells. Targeted deletion of IL-13 in mice resulted in impaired Th2 cell development and indicated an important role for IL-13 in the expulsion of gastrointestinal parasites. IL-13 exerts anti-inflammatory effects on monocytes and macrophages and it inhibits the expression of inflammatory cytokines such as IL-1β, TNF-α, IL-6 and IL-8. IL-13 has also been shown to enhance B cell proliferation and to induce isotype switching resulting in increased production of IgE. Blocking of IL-13 activity inhibits the pathophysiology of asthma. Human and murine IL-13 is cross-species reactive. A variant of IL-13 shows enhanced functional activity compared with the wild type IL-13. This genetic variant, termed human IL-13 analog, is a mature 114 amino acid protein with a substitution of Q for R at position 112.

sRANK Ligand Antibody: The receptor activator of NF-kappa B ligand (RANK-L) is a recently discovered member of the TNF-ligand family involved in the regulation of the T cell-dependent immune response, lymph node organogenesis and bone formation. RANK-L exists as both a normal, transmembrane form and a truncated, soluble form (sRANK-L), both of which can stimulate the receptor. Activation of T cells, such as by treatment with interleukin-7, induces RANK-L production and leads to an increase of osteoclast formation and bone loss. Finally, sRANK-L can activate the antiapoptotic kinase Akt through a signaling complex involving Src kinase and TRAF6, suggesting sRANK-L may also play a role in regulating apoptosis. This antibody will recognize both the soluble form and the uncleaved transmembrane form of RANK-L.
