T cells require a signal induced by the engagement of the T cell receptor and a costimulatory signal(s) through distinct T cell surface molecules for optimal T cell activation and tolerance. CD273 (PD-L2) is one of two ligands for programmed death-1 (PD-1; CD279), a member of the CD28 family of immunoreceptors. The other identified ligand is PD-L1. CD273 is broadly expressed and also up regulated in a variety of tumor cell lines. On previously activated T cells, CD273 interaction with PD-1 inhibits TCR mediated proliferation and cytokine production, suggesting an inhibitory role in regulating immune responses. CD273 has a costimulatory function on resting T cells activated with suboptimal TCR signals.

This GLP-1 (7-36) amide, is labeled with biotin at the peptide N-terminus. GLP-1 (7-36) amide is an incretin hormone that causes glucose dependent release of insulin by pancreatic beta cells. It is the cleavage product of GLP-1 (1-36) amide peptide (Cat# AS-22460). Both GLP-1 (7-36) and GLP-1 (7-37)-Cat# AS-20761, also play roles in gastric motility (gastric emptying), on the suppression of plasma glucagon levels (glucose production) and possibly on the promotion of satiety and stimulation of glucose disposal in peripheral tissues independent of the actions of insulin. GLP-1 (7-36) has a short half life of less than 2 minutes, and like GIP, is rapidly degraded by the enzyme dipeptidyl peptidase IV (DPP-4), which is widely expressed in a number of sites, including the endothelial cells of small gut arterioles. DPP-4 degrades GLP-1 (7-36) into the non insulinotropic GLP-1 (9-36)-Cat# AS-65070 (some studies suggest it may have weak insulinotropic activity). As a result, the majority of GLP-1 (and GIP) is inactivated as an insulinotrope before reaching the systemic circulation.

Interleukin-27 (IL-27) is a heterodimeric group 2 receptor ligand molecule that belongs to the IL-6/IL-12 family of long type I cytokines. It is composed of EBI3 (EBV-induced gene 3), a 34 kDa glycoprotein that is related to the p40 subunit of IL-12 and IL-23, and p28, the cloned 28 kDa glycoprotein that is related to the p35 chain of IL-12. IL-27 is expressed by monocytes, endothelial cells and dendritic cells. IL-27 binds to and signals through a heterodimeric receptor complex composed of WSX1 (TCCR) and gp130. Evidence suggests IL-27 interacts only with WSX-1. IL-27 has both anti- and proinflammatory properties. As an antiinflammatory, IL-27 seems to induce a general negative feedback program that limits T and NK-T cell activity. At the onset of infection, IL-27 induces an IL-12 receptor on naïe CD4+ T cells, making them susceptible to subsequent IL-12 activity (and possible Th1 development). Notably, IL-12 family cytokines are both induced and inhibited by bacterial products. Microbes promote IL-27 secretion through TLR4, and also block IL-27 production via C5a induction.

FAM36A is a multi-pass membrane protein. It belongs to the FAM36 family. The exact function of FAM36A remains unknown.Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The FAM36A gene product has been provisionally designated FAM36A pending further characterization.

HETEs are byproducts generated by the metabolism of arachidonic acid by lipoxygenases. 12(S)-HETE is the stereospecific hydroxy product from the reduction of 12(S)-hydroperoxy tetraenoic eicosatetraenoic acid [12(S)-HpETE], which itself is a 12-lipoxygenase metabolite of arachidonic acid. 12(S)-HETE is chemotactic and chemokinetic for polymorphonuclear leukocytes and vascular smooth muscle cells. It also acts as a second messenger in angiotensin-II induced aldosterone production. 15-hydroxyeicosatetraenoic acid [15(S)-HETE] is the major hydroxy derivative of arachidonic acid when acted upon by 15-lipoxygenase. It is also the primary monohydroxy acid synthesized by the lipoxygenase activity of Cyclooxygenase-1 upon linoleic acid. 15(S)-HETE acts as a paracrine regulator of smooth muscle and lung neutrophil recruitment, and elevated levels of 15(S)-HETE are associated with asthma, rhinitis, chronic paranasal sinusitis and rheumatoid arthritis.

Interleukin-17 is a potent pro-inflammatory cytokine produced by activated memory T cells. There are at least six members of the IL-17 family in humans and in mice. As IL-17 shares properties with IL-1 and TNF-alpha, it may induce joint inflammation and bone and cartilage destruction. This cytokine is found in synovial fluids of patients with rheumatoid arthritis, and produced by rheumatoid arthritis synovium. It increases IL-6 production, induces collagen degradation and decreases collagen synthesis by synovium and cartilage and proteoglycan synthesis in cartilage. IL-17 is also able to increase bone destruction and reduce its formation. Blocking of interleukin-17 with specific inhibitors provides a protective inhibition of cartilage and bone degradation.
