Interleukin 12 (IL-12) is an inducer of cell-mediated immunity and is produced by dendritic cells, macrophages, and B cells after antigen stimulation. IL-12 functions to promote naïve T cell differentiation into the type 1 T helper cell (Th1) lineage. The protein encoded by this gene is a multifunctional, nuclear phosphoprotein that plays a role in cell cycle progression, apoptosis and cellular transformation. It functions as a transcription factor that regulates transcription of specific target genes. Mutations, overexpression, rearrangement and translocation of this gene have been associated with a variety of hematopoietic tumors, leukemias and lymphomas, including Burkitt lymphoma. There is evidence to show that alternative translation initiations from an upstream, in-frame non-AUG (CUG) and a downstream AUG start site result in the production of two isoforms with distinct N-termini. Interleukin-18-binding protein (IL18BP) is also known as Tadekinig-alfa, IL18BPa, which functions as an IL18 inhibitor. IL18BP is a secreted protein which contains one Ig-like C2-type (immunoglobulin-like) domain. IL18BP does not have a transmembrane domain and hence is not anchored to the cell membrane. IL18BP is strongly expressed in heart, lung, placenta and spleen. Isoform A of IL18BP binds to IL-18 and inhibits its activity. IL18BP also functions as an inhibitor of the early TH1 cytokine response. IL18BP suppresses the production of IFN-gamma resulting in reduced T-helper type 1 immune responses.
