TBX1 is a member of a phylogenetically conserved family of genes that share a common DNA-binding domain, the T-box. T-box genes encode transcription factors involved in the regulation of developmental processes. TBX1 product shares 98% amino acid sequence identity with the mouse ortholog. DiGeorge syndrome (DGS)/velocardiofacial syndrome (VCFS), a common congenital disorder characterized by neural-crest-related developmental defects, has been associated with deletions of chromosome 22q11.2, where TBX1 has been mapped. Studies using mouse models of DiGeorge syndrome suggest a major role for this gene in the molecular etiology of DGS/VCFS. Several alternatively spliced transcript variants encoding different isoforms have been described for this gene.

Toll-Interacting Protein (TOLLIP) is a member of the tollip family. TOLLIP localizes to the cytoplasm. It contains one C2 domain and one CUE domain. TOLLIP is an inhibitory adaptor protein for Toll-like receptors (TLR). The Toll-like receptors pathway is a part of the immune system that recognize structurally conserved molecular patterns of microbial pathogens, resulting in an inflammatory immune response. TOLLIP constitutes a complex with Tom1 to regulate endosomal transferring of ubiquitinated proteins. TOLLIP can negative regulate Toll-like receptors signaling, which may limit the production of proinflammatory mediators during the process of inflammation and infection.

The protein encoded by this gene is a member of the lipophilin subfamily, part of the uteroglobin superfamily, and is an ortholog of prostatein, the major secretory glycoprotein of the rat ventral prostate gland. Lipophilin gene products are widely expressed in normal tissues, especially in endocrine-responsive organs. Assuming that human lipophilins are the functional counterparts of prostatein, they may be transcriptionally regulated by steroid hormones, with the ability to bind androgens, other steroids and possibly bind and concentrate estramustine, a chemotherapeutic agent widely used for prostate cancer. Although the gene has been reported to be on chromosome 10, this sequence appears to be from a cluster of genes on chromosome 11 that includes mammaglobin 2.
