This protein is found to possess 3' to 5' exonuclease activity, which may contribute to its role in sensing and repairing DNA damage. It forms a checkpoint protein complex with RAD1 and HUS1. This complex is recruited by checkpoint protein RAD17 to the sites of DNA damage, which is thought to be important for triggering the checkpoint-signaling cascade. Representing about 2% of human DNA, chromosome 20 consists of approximately 63 million bases and 600 genes. Chromosome 20 contains a region with numerous genes expressed in the epididymis which are thought important for seminal production and some viewed as potential targets for male contraception. The PRNP gene encoding the prion protein associated with spongiform encephalopathies, like Creutzfeldt-Jakob disease, is found on chromosome 20. Amyotrophic lateral sclerosis, spinal muscular atrophy, ring chromosome 20 epilepsy syndrome and Alagille syndrome are also associated with chromosome 20.

The product of this gene functions in the NF-kappaB signal transduction pathway. The encoded protein, and the similar and functionally redundant protein MAP3K7IP2/TAB2, forms a ternary complex with the protein kinase MAP3K7/TAK1 and either TRAF2 or TRAF6 in response to stimulation with the pro-inflammatory cytokines TNF or IL-1. Subsequent MAP3K7/TAK1 kinase activity triggers a signaling cascade leading to activation of the NF-kappaB transcription factor. The human genome contains a related pseudogene. Alternatively spliced transcript variants have been described, but their biological validity has not been determined.

The Olfactomedin family comprises a diverse group of secreted glycoproteins, which includes OLFM1 (Noelin-1), OLFM2 (Noelin-2), OLFM3 (Noelin-3), OLFM4 (Noelin-4), tiarin, pancortin, gliomedin and mycocilin. These proteins are implicated in the development of the nervous system. Specifically, OLFM1 and OLFM2 expression is observed in the neural plate and neural crest, as well as in the cranial ganglia in mouse at E8-10, and later in brain tissue and in the zone of polarizing activity in the limb. Overexpression of OLFM1 causes an excess of neural crest emigrations and prolonged neural crest production. OLFM2 participates in the regulation of the development of the anterior nervous system. An Arg144Gln mutation in OLFM2 has been implicated as a possible cause for open-angle glaucoma (OAG).

CESK1, also known as CCT8L2 (chaperonin containing TCP1, subunit 8 theta-like 2), is a 557 amino acid protein that localizes to the cytoplasm and is thought to function as a molecular chaperone, possibly assisting protein folding after ATP hydrolysis. CESK1 belongs to the TCP-1 chaperonin family and is encoded by a gene which maps to human chromosome 22. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, neurofibromatosis type 2, autism and schizophrenia. Additionally, translocations between chromosomes 9 and 22 may lead to the formation of the Philadelphia chromosome and the subsequent production of the novel fusion protein Bcr-Abl, a potent cell proliferation activator found in several types of leukemias.
