The product of this gene belongs to a subfamily of calcium binding proteins, which share similarity to calmodulin. Calcium binding proteins are an important component of calcium mediated cellular signal transduction. Expression of this gene was only detected in retina and brain. Study of the mouse homolog demonstrated that groups of cells expressing this protein are located in the center or inner border of the inner unclear layer of retina. Three alternatively spliced variants encoding different isoforms have been described.

This antibody reacts with a 75 kDa melanocyte-specific gene product, identified as Tyrosinase-related protein-1 (TRP-1). It is involved in melanin synthesis. TRP1 is present on the melanosomal membranes of melanoma, normal melanocytes and nevi.Recent evidence suggests that TRP-1 is involved in maintaining stability of tyrosinase protein and modulating its catalytic activity. TRP-1 is also involved in maintenance of melanosome ultrastructure and affects melanocyte proliferation and cell death.

The protein encoded by this gene is a small GTPase of the Rho-subfamily, which regulates signaling pathways that control diverse cellular functions including cell morphology, migration, endocytosis and cell cycle progression. This protein is highly similar to Saccharomyces cerevisiae Cdc 42, and is able to complement the yeast cdc42-1 mutant. The product of oncogene Dbl was reported to specifically catalyze the dissociation of GDP from this protein. This protein could regulate actin polymerization through its direct binding to Neural Wiskott-Aldrich syndrome protein (N-WASP), which subsequently activates Arp2/3 complex. Alternative splicing of this gene results in multiple transcript variants. Pseudogenes of this gene have been identified on chromosomes 3, 4, 5, 7, 8 and 20.

MSI2 Antibody: Musashi2 (MSI2) is an RNA-binding protein that is highly expressed in precursor cells in the ventricular and subventricular zones of the developing mammalian CNS. Like the related MSI1, MSI2 has been suggested to be involved stem cell production and maintenance. MSI2 is the predominant MSI protein in hematopoietic stem cells, and its knockdown leads to reduced engraftment and depletion in vivo. Expression levels of MSI2 are elevated in myeloid leukemia cells lines, and MSI2 appears to cooperate with BCR-ABL1 to induce an aggressive leukemia; the level of MSI2 directly correlates with decreased survival in patients. MSI2 negatively regulates the asymmetric cell fate determinant NUMB, suggesting that this signaling pathway may provide future targets for future therapies.

OTUD5 Antibody: OTUD5 is a member of the OTU (ovarian tumor) domain-containing cysteine protease superfamily. The OTU domain confers deubiquitinase activity and OTUD5 has been shown to suppress the type I interferon (IFN-I)-dependent innate immune response by cleaving the polyubiquitin chain from TRAF3, an essential type I interferon adaptor protein. Cleavage results in disassociation of TRAF3 from a downstream signaling complex containing TBK1 and the disruption of the IFN-I signaling cascade, indicating that OTUD5 acts as a negative regulator of innate immune responses. It has been suggested that by suppressing IFN-I production, OTUD5 may function to inhibit the emergence of certain autoimmune disorders such as systemic lupus erythematosus. Multiple isoforms of OTUD5 are known to exist.

IL-23 Antibody: Like interleukin-27 (IL-27), IL-23 is a recently discovered member of the IL-6/IL-12 family of proinflammatory and immunoregulatory cytokines. It exists as a heterodimer composed of the IL-12p40 subunit and a novel p19 subunit. IL-23 is secreted by activated dendritic cells, macrophages, and monocytes. Its biological activities include enhancing the proliferation of memory T cells and the production of IFN-gamma, IL-12, and TNF-alpha from activated T cells, and can stimulate macrophages to produce TNF-alpha and nitric oxide. It has also been shown to possess potent anti-tumor and anti-metastatic activity in mouse models of cancer, suggesting a potential role for IL-23 in therapeutic treatment of cancer.

SLAMF3 Antibody: The signaling lymphocyte-activation molecule family member 3 (SLAMF3), also known as LY9, is a cell surface receptor that is expressed on T and B lymphocytes and belongs to the CD150/SLAMF1 receptor family. SLAMF3 was identified through a yeast two-hybrid screening in which SLAMF3 bound to the X-linked lymphoproliferative disease gene product SAP, suggesting that in activated T cells, SLAMF3 signaling can be triggered SAP. Its expression on the cell surface appears to be regulated via its interaction with the clathrin-associated adaptor complex 2 (AP-2). Despite its similarity to SLAMF1 in structure and interactions with SAP, SLAMF3-deficient mice do not exhibit phenotypic characteristics associated with SLAMF1- and SAP-deficient mice, suggesting that SLAMF3 plays other roles in T cell activation.

Interleukin-22 (IL-22), also known as IL-10 related T cell derived inducible factor (ILTIF) was initially identified as a gene induced by IL-9 in mouse T cells and mast cells. IL-22 has been shown to activate STAT1 and STAT3 in several hepatoma cell lines and upregulate the production of acute phase proteins. IL-22 is produced by normal T cells upon anti-CD3 stimulation in humans. Mouse IL-22 expression is also induced in various organs upon lipopolysaccharide injection, suggesting that IL-22 may be involved in inflammatory responses. The functional IL-22 receptor complex consists of two receptor subunits, IL-22R(CRF29) and IL-10Rbeta(CRF24), belonging to the class II cytokine receptor family.
