The small leucine-rich proteoglycan (SLRP) family of proteins contains various proteins such as Decorin, Biglycan, Fibromodulin, Keratocan, Lumican, Osteoadherin and Osteoglycin. These proteins all have similar functions as they all mediate extracellular matrix organization and act as binding partners of TGF beta. Osteoglycin, which also may be designated osteoinductive factor (OIF), is a secreted protein detected in bone tissues. Osteoglycin induces the formation of bone in conjunction with either TGF-beta-1 or TGF-beta-2. The precursor form of the OGN gene product, designated Mimecan, is subject to in situ proteolytic cleavage to yield the mature Osteoglycin.

FNIP2 Antibody: FNIP2 is the second protein found to interact with folliculin, the product of the Birt-Hogg-Dube (BHD) gene. Folliculin is thought to act as a tumor suppressor as mutations or loss of heterozygosity in this gene are associated with BHD syndrome-related renal tumors. Folliculin and FNIP1, a protein that shares 49% identity to FNIP2, bind to AMPK, an important energy sensor in cells that negatively regulates the mammalian target of rapamycin (mTOR), a protein that is thought to be the master switch for cell growth and proliferation. FNIP1 and FNIP2 are able to form homo- and heteromeric multimers, suggesting these proteins may have a functional relationship.

APH1 Antibody: APH1 was initially identified as a component of the Notch pathway in C. elegans. Along with nicastrin, PEN2, and presenilin-1 APH1 is an essential component of the gamma-secretase complex which cleave the amyloid precursor protein (APP) at what are known as the gamma- and epsilon-sites and can lead to the accumulation of the Amyloid beta peptide (Abeta) cleavage product that is associated with Alzheimer's disease. APH1 exists in at least three distinct isoforms with APH1a as the principal isoform present in the gamma-secretase complex. Mice deficient in this isoform, but not the other two, were lethal at E10.5, with impaired vascular and neural development observed.

MIF (macrophage migration inhibitory factor) was one of the first cytokine activities to be discovered and was initially described as a T cell-derived factor that inhibits the random migration of macrophages (Weiser 1989). Recently, MIF was rediscovered as a pituitary hormone that act as the counterregulatory hormone for glucocorticoid action within the immune system (Bernhagen 1993, Mitchell 1995). MIF was released from macrophages and T cells in response to physiological concentrations of glucocorticoids. The secreted MIF counter-regulates the immunosuppressive effects of steroids on immune cell activation and cytokine production (Bucala 1998). MIF plays a critical role in the host control of inflammation and immunity.

The protein encoded by this gene is a guanine nucleotide exchange factor (GEF) similar to the Saccharomyces cerevisiae CDC25 gene product. Functional analysis has demonstrated that this protein stimulates the dissociation of GDP from RAS protein. The studies of the similar gene in mouse suggested that the Ras-GEF activity of this protein in brain can be activated by Ca2+ influx, muscarinic receptors, and G protein beta-gamma subunit. Mouse studies also indicated that the Ras-GEF signaling pathway mediated by this protein may be important for long-term memory. Alternatively spliced transcript variants encoding distinct isoforms have been reported.

These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that they either directly regulate gamma-secretase activity or themselves are protease enzymes. TIM-1 Antibody: The human form of TIM-1 was initially discovered as a membrane glycoprotein through which the hepatitis A virus can gain entry into a cell. It was also identified as kidney injury molecule 1 (Kim-1), a predicted adhesion molecule that is upregulated on the surfaces of kidney epithelia. It is also expressed on T helper 2 (Th2) cells of the immune system, and following the binding of its natural ligand TIM-4, stimulates T cell expansion and cytokine production in response to viral challenge. It has been suggested that hyperactivation of TIM-1 leads to an increased level of Th2 responsiveness and asthma susceptibility, and antibodies to TIM-1 may therefore be a novel approach to treating asthma.
