Alzheimer's disease (AD) patients with an inherited form of the disease carry mutations in the presenilin proteins (PSEN1;PSEN2) or the amyloid precursor protein (APP). These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that they either directly regulate gamma-secretase activity or themselves are protease enzymes. Three alternative splice variants of PSEN1 have been identified.

Caspase 12 is an enzyme known as a cysteine protease. It belongs to a family of enzymes called caspases that cleave their substrates at C-terminal aspartic acid residues. It is most highly related to members of the ICE subfamily of caspases that process inflammatory cytokines. Caspase-12 is located in the endoplasmic reticulum (ER). It is responsible for ER-stress-induced apoptosis, such as high calcium concentration, low oxygen and low glucose levels. Caspase-12 Antibody detects endogenous levels of full-length caspase-12 protein (50 kDa) and its cleaved product (40-44 kDa). This antibody does not cross-react with other caspases.

TIM-1 Antibody: The human form of TIM-1 was initially discovered as a membrane glycoprotein through which the hepatitis A virus can gain entry into a cell. It was also identified as kidney injury molecule 1 (Kim-1), a predicted adhesion molecule that is upregulated on the surfaces of kidney epithelia. It is also expressed on T helper 2 (Th2) cells of the immune system, and following the binding of its natural ligand TIM-4, stimulates T cell expansion and cytokine production in response to viral challenge. It has been suggested that hyperactivation of TIM-1 leads to an increased level of Th2 responsiveness and asthma susceptibility, and antibodies to TIM-1 may therefore be a novel approach to treating asthma.

The EBVCS-5 antibody is specific for human CD23, also known as the low affinity IgE receptor (FceRII), a 45 kDa type II transmembrane protein and member of the immunoglobulin superfamily. It is expressed on most B cells and is upregulated upon activation. It is also present on mantle zone B cells, eosinophils, monocytes and a subset of T cells and platelets. CD23 plays a role in B cell development and differentiation, and also functions to regulate IgE production. MIF (macrophage migration inhibitory factor) was one of the first cytokine activities to be discovered and was initially described as a T cell-derived factor that inhibits the random migration of macrophages (Weiser 1989). Recently, MIF was rediscovered as a pituitary hormone that act as the counterregulatory hormone for glucocorticoid action within the immune system (Bernhagen 1993, Mitchell 1995). MIF was released from macrophages and T cells in response to physiological concentrations of glucocorticoids. The secreted MIF counter-regulates the immunosuppressive effects of steroids on immune cell activation and cytokine production (Bucala 1998). MIF plays a critical role in the host control of inflammation and immunity.

These proteins all have similar functions as they all mediate extracellular matrix organization and act as binding partners of TGF beta. Osteoglycin, which also may be designated osteoinductive factor (OIF), is a secreted protein detected in bone tissues. Osteoglycin induces the formation of bone in conjunction with either TGF-beta-1 or TGF-beta-2. Along with nicastrin, PEN2, and presenilin-1 APH1 is an essential component of the gamma-secretase complex which cleave the amyloid precursor protein (APP) at what are known as the gamma- and epsilon-sites and can lead to the accumulation of the Amyloid beta peptide (Abeta) cleavage product that is associated with Alzheimer's disease. APH1 exists in at least three distinct isoforms with APH1a as the principal isoform present in the gamma-secretase complex. Mice deficient in this isoform, but not the other two, were lethal at E10.5, with impaired vascular and neural development observed.
