Interleukin (IL)-10 is an anti-inflammatory cytokine, produced by T helper (Th) cells, macrophages, monocytes, and B cells, that plays a crucial role in preventing inflammatory and autoimmune pathologies. It downregulates the expression of Th1 cytokines, MHC class II antigens, and co-stimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. IL-10 can block NF-κB activity, and is involved in the regulation of the JAK-STAT signaling pathway. IL-10, along with its receptors, describes an important role in pathogenesis of various diseases, including infectious, inflammatory, autoimmune diseases. IL-10 mutations are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis.

Two retinoblastoma related proteins, p107 and pRb2 / p130, which are structurally and functionally similar to the product of the retinoblastoma gene (pRb / p105), were cloned by taking advantage of their ability to bind transforming proteins of DNA tumor viruses through a particular region called the "pocket domain." Like pRb, both proteins play a fundamental role in growth control. These Rb family proteins were measured in a variety of lung neoplasms. The highest percentage of undetectable levels and the tightest inverse correlation with the histological grading and with PCNA expression in the most aggressive tumor types were found for pRb2 / p130, which may suggest an important role for this protein in the pathogenesis and progression of lung cancer.

Anti-PNL2 is a novel monoclonal antibody, which has recently been introduced as an immunohistochemical reagent to stain melanocytes and tumors derived therefrom. The antigen recognized by PNL2 is different from Melan A and gp100. Its epitope is not destroyed by digestion with neuraminidase i.e. its epitope id not glycosylated. Anti-PNL2 may be most useful because of its high sensitivity for metastatic melanoma (87%), as opposed to 76% for anti-HMB45 and 82% for anti-MART-1. Anti-PNL2 labels intra-epidermal nevi while the dermal component of compound nevi are largely non-reactive with anti-PNL2. Antibodies against PNL2, MART-1 (Melan A) and HMB45 stain most clear cell sarcoma cells and a few cells in angio-myolipomas and lymphangioleiomyomatosis. Anti-PNL2 is a useful antibody for the identification of melanomas and clear cell sarcomas. Differential diagnosis is aided by the results from a panel of antibodies, including antibodies against HMB45, MART-1, tyrosinase, and MiTF.
