Mature Myostatin is obtained by proteolytic processing of a biologically-inactive precursor protein, which contains an N-terminal propeptide of 243 amino acid residues. Myostatin-Propeptide exhibits high binding affinity for myostatin, and has been shown to be a potent inhibitor of myostatin. Over-expression of myostatin-propeptide in mice resulted in large increases (up to 200%) in skeletal muscle mass, similar to those observed in myostatin knockout mice. Recombinant Human Myostatin-Propeptide is a 27.8 kDa protein consisting of 244 amino acid residues.

Osteoprotegerin (OPG, or osteoclastogenesis inhibitory factor) is a secretory glycoprotein belonging to TNF receptor superfamily. Acts as decoy receptor for RANKL and thereby neutralizes its function in osteoclastogenesis. Inhibits the activation of osteoclasts and promotes osteoclast apoptosis. Bone homeostasis seems to depend on the local RANKL/OPG ratio. May also play a role in preventing arterial calcification. May act as decoy receptor for TRAIL and protect against apoptosis. TRAIL binding blocks the inhibition of osteoclastogenesis. OPG acts as a soluble factor in the regulation of bone mass and may be beneficial in the treatment of osteoporosis with increased osteoclast activity. Defects in CYLD are the cause of familial cylindromatosis (CYLD) also known as turban tumor syndrome or dermal eccrine cylindromatosis. CYLD is an autosomal dominant and highly tumor type-specific disorder. The tumors (known as cylindromas because of their characteristic microscopic architecture) are believed to arise from or recapitulate the appearance of the eccrine or apocrine cells of the skin that secrete sweat and scent respectively. Cylindromas arise predominantly in hairy parts of the body with approximately 90% on the head and neck. The development of a confluent mass which may ulcerate or become infected has led to the designation "turban tumor syndrome". The skin tumors show differentiation in the direction of hair structures, hence the synonym trichoepithelioma. CYLD has deubiquitinating activity.
