This MAb recognizes a single chain, transmembrane, heavily glycosylated protein of 90-120 kDa, which is identified as CD34. Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, CD34 is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. Its expression is also found in vascular endothelium. Additionally, it appears that proliferating endothelial cells express this molecule more than the non-proliferating endothelial cells. Anti-CD34 labels > 85% of angiosarcoma and Kaposi's sarcoma, but with a lower specificity.

Description:
CD31 (PECAM-1) is a transmembrane glycoprotein member of the immunoglobulin supergene family of adhesion molecules. CD31 is expressed by stem cells of the hematopoietic system and is primarily used to identify and concentrate these cells for experimental studies as well as for bone marrow transplantation. Anti-CD31 has shown to be highly specific and sensitive for vascular endothelial cells. Staining of nonvascular tumors (excluding hematopoietic neoplasms) is rare. CD31 MAb reacts with normal, benign, and malignant endothelial cells which make up blood vessel lining. The level of CD31 expression can help to determine the degree of tumor angiogenesis, and a high level of CD31 expression may imply a rapidly growing tumor and potentially a predictor of tumor recurrence.

This antibody reacts with Ig domain of CD47 protein. It has been shown to inhibit polymorphonuclear neutrophil (PMN) transmigration across cell monolayers and matrix. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. CD47 may play a role in membrane transport and/or integrin dependent signal transduction.

This MAb recognizes a 60 kDa antigen associated with the mitochondria in cells. It is a part of a new panel of reagents, which recognizes subcellular organelles or compartments of cells. These markers may be useful in identification of these organelles in cells, tissues, and biochemical preparations. It recognizes an antigen associated with the mitochondria in cells from a wide variety of animals, but not insects and bacteria. It can be used to stain the mitochondria in cell or tissue preparations and can be used as a mitochondrial marker in subcellular fractions. It produces a spaghetti-like pattern in normal and malignant cells and may be used to stain mitochondria of cells in fixed or frozen tissue sections. It can also be used with paraformaldehyde fixed frozen tissue or cell preparations.

This antibody recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO). This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.
