Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

CF®405S is a blue fluorescent dye (Ex/Em 404/431 nm) with superior brightness compared to other blue dyes; it is also compatible with super-resolution imaging by SIM. This antibody recognizes a protein of ~55 kDa, identified as SOX10. This MAb is highly specific and does not cross-react with other members of the SOX-family. SOX genes comprise a family of genes that are related to the mammalian sex-determining gene SRY. These genes similarly contain sequences that encode for the HMG-box domain, which is responsible for the sequence-specific DNA-binding activity. SOX-10 is a sensitive marker of melanoma, including conventional, spindled, and desmoplastic subtypes. It is expressed by metastatic melanomas and nodal capsular nevus in sentinel lymph nodes, but not by other lymph node components such as dendritic cells, which usually express S100 protein. Commonly used melanoma markers, such as anti-HMB-45 and anti-Melan-A, are poorly expressed in desmoplastic melanomas while SOX-10 is moderately to strongly expressed in desmoplastic melanomas. SOX-10 is considered as a very reliable marker for recognizing residual desmoplastic melanomas. In normal tissues, it is expressed in Schwann cells, melanocytes, and myoepithelial cells of salivary, bronchial and mammary glands. SOX-10 expression is also observed in mast cells.

This antibody reacts with tissue non-specific alkaline phosphatase. There are at least four distinct but related alkaline phosphatases: intestinal, placental, placental-like, and liver/bone/kidney (tissue non-specific). The first three are located together on chromosome 2, while the tissue non-specific form is located on chromosome 1. The product of this gene is a membrane bound glycosylated enzyme that is not expressed in any particular tissue and is, therefore, referred to as the tissue-nonspecific form of the enzyme. The exact physiological function of the alkaline phosphatases is not known. A proposed function of this form of the enzyme is matrix mineralization; however, mice that lack a functional form of this enzyme show normal skeletal development. This enzyme has been linked directly to hypo-phosphatasia, a disorder that is characterized by hypercalcemia and includes skeletal defects. The character of this disorder can vary, however, depending on the specific mutation since this determines age of onset and severity of symptoms. Alternatively spliced transcript variants, which encode the same protein, have been identified for this gene.

Mucin 5AC glycoprotein (MUC5AC) is a 641 kDa glycoprotein belonging to the superfamily of mucins. Mucins are high molecular weight glycoproteins produced by epithelial cells and can be divided into two families; secretory mucins and membrane bound mucins. MUC5AC is a mucus-forming secreted mucin that is found in normal gastric and tracheo-bronchial mucosa, but absent from normal colon. MUC5AC expression is present in primary ovarian mucinous cancer but usually absent in colorectal adenocarcinoma, thus showing an expression pattern opposite to MUC2. Together with a panel of antibodies, Anti-MUC5AC may be useful for differential identification of primary mucinous ovarian tumors from colon adenocarcinoma metastatic to the ovary. MUC5AC antibodies may also be useful for identification of intestinal metaplasia as well as in the identification of pancreatic carcinoma and pre-cancerous changes vs. normal pancreas.

Description:
Actin is a major component of the cytoskeleton and is present in most cell types. It is highly specific to actin from smooth muscles. This MAb does not stain cardiac or skeletal muscle; however, it does stain myofibroblasts and myoepithelial cells. This antibody could be used together with anti-muscle specific actin and myogenin in making a diagnosis of smooth muscle and skeletal muscle tumors. In most cases of rhabdomyosarcoma, this antibody yields negative results whereas anti-muscle specific actin and myogenin are positive. Leiomyosarcomas are positive only with anti-muscle specific actin and anti-smooth muscle actin and are negative with anti-myogenin.

Podocalyxin is a member of the CD34 transmembrane sialomucin family. It is over-expressed on the podocyte foot projections and plays essential roles in kidney development and homeostasis, blood filtration and urine formation. It is also expressed on vascular endothelia, hematopoietic progenitors and a subset of neurons. Overexpression of podocalyxin may be linked to more aggressive tumor behavior. Podocalyxin antibody can identify podocytes in the urine (podocyturia) that may indicate glomerular disease, pre-eclampsia, and other kidney pathology.
